Defects in the NC2 repressor affect both canonical and non-coding RNA polymerase II transcription initiation in yeast.
2016
BACKGROUND: The formation of the pre-initiation complex in eukaryotic genes is a key step in transcription initiation. The TATA-binding protein (TBP) is a universal component of all pre-initiation complexes for all kinds of RNA polymerase II (RNA pol II) genes, including those with a TATA or a TATA-like element, both those that encode proteins and those that transcribe non-coding RNAs. Mot1 and the negative cofactor 2 (NC2) complex are regulators of TBP, and it has been shown that depletion of these factors in yeast leads to defects in the control of transcription initiation that alter cryptic transcription levels in selected yeast loci. RESULTS: In order to cast light on the molecular func…
Serological Follow-Up Study Indicates High Seasonal Coronavirus Infection and Reinfection Rates in Early Childhood
2022
Seasonal human coronaviruses (HCoVs) cause respiratory infections, especially in children. Currently, the knowledge on early childhood seasonal coronavirus infections and the duration of antibody levels following the first infections is limited. Here we analyzed serological follow-up samples to estimate the rate of primary infection and reinfection(s) caused by seasonal coronaviruses in early childhood. Serum specimens were collected from 140 children at ages of 13, 24, and 36 months (1, 2, and 3 years), and IgG antibody levels against recombinant HCoV nucleoproteins (N) were measured by enzyme immunoassay (EIA). Altogether, 84% (118/140) of the children were seropositive for at least one s…
A conspectus of Tephroseris (Asteraceae: Senecioneae) in Europe outside Russia and notes on the decline of the genus
2021
Tephroseris is generally considered a difficult genus. Based on the examination of extensive herbarium material and considering the existing literature, we recognize seven species in Europe outside Russia. These are T. palustris, T. integrifolia with subsp. integrifolia, subsp. aurantiaca, subsp. capitata, subsp. maritima, subsp. serpentini and subsp. “tundricola”, T. balbisiana, T. crispa, T. helenitis, T. longifolia and T. papposa. Phylogenetic analysis of ITS and ETS sequences showed that these species fall into three lineages. These are: (1) T. palustris, clearly related to Arctic species of the genus; (2) T. integrifolia; and (3) the remaining species. Molecular dating of the T. integr…
Effects ofBacillus thuringiensisCry1Ab and Cry3Aa endotoxins on predatory Coleoptera tested through artificial diet-incorporation bioassays
2009
AbstractTraditional approaches to studying the effects of genetically modified (GM) crops on beneficial insects involve either field assays, comparing insect population levels between control and GM crops or tritrophic bioassays with contaminated insects – usually larvae or eggs of Lepidoptera – as preys. Here, we report the results of a bioassay using an artificial diet, suitable for predatory Coleoptera, to supplyBacillus thuringiensis(Bt) solubilized Cry1Ab and Cry3Aa as well as trypsin-activated Cry1Ab toAtheta coriariaandCryptolaemus montrouzieriadults and young larvae ofAdalia bipunctata. Water, solubilization buffer and trypsin-treated solubilization buffer were used as controls. In …
Phenotype associated with TAF2 biallelic mutations: a clinical description of four individuals and review of the literature
2021
International audience; Transcription factor IID is a multimeric protein complex that is essential for the initiation of transcription by RNA polymerase II. One of its critical components, the TATA-binding protein-associated factor 2, is encoded by the gene TAF2. Pathogenic variants of this gene have been shown to be responsible for the Mental retardation, autosomal recessive 40 syndrome. This syndrome is characterized by severe intellectual disability, postnatal microcephaly, pyramidal signs and thin corpus callosum. Until now, only three families have been reported separately. Here we report four individuals, from two unrelated families, who present with severe intellectual disability and…
Posterior tibial tendon dysfunction: Clinical and magnetic resonance imaging findings having histology as reference standard.
2017
Abstract Objective To investigate the correlation between MRI, clinical tests, histopathologic features of posterior tibial tendon (PTT) dysfunction in patients with acquired adult flatfoot deformity surgically treated with medializing calcaneal osteotomy and flexor digitorum longus tendon transposition. Materials and methods Nineteen patients (11 females; age: 46 ± 15 year, range 18–75) were pre-operatively evaluated using the single heel rise (HR) and the first metatarsal rise (FMR) sign tests. Two reviewers graded the PTT tears on a I–III scale and measured the hindfoot valgus angle on the pre-operative MRI of the ankle. The specimens of the removed portion of PTT were histologically ana…
Effect of static foot posture on the dynamic stiffness of foot joints during walking
2018
Background The static foot posture has been related to the development of lower limb injuries. Research question This study aimed to investigate the dynamic stiffness of foot joints during gait in the sagittal plane to understand the role of the static foot posture in the development of injuries. Methods Seventy healthy adult male subjects with different static postures, assessed by the Foot Posture Index (FPI) (30 normal, 20 highly pronated and 20 highly supinated), were recruited. Kinematic and kinetic data were recorded using an optical motion capture system and a pressure platform, and dynamic stiffness at the different stages of the stance was calculated from the slopes of the linear r…
Dynamic Flexion Stiffness of Foot Joints During Walking
2016
Background: Dynamic stiffness can be used for studying foot pathologic abnormalities and for developing prostheses and orthoses. Although previous works have studied the role of ankle joint stiffness during gait, other foot joints have not yet been analyzed. We sought to characterize the dynamic stiffness of the ankle, midtarsal, and metatarsophalangeal joints during normal walking. Methods: Kinematics and contact data from four healthy individuals during walking were registered with a three-dimensional motion analysis system and a pressure platform. Stance phases with flexion moment-angle linear relationships were identified, and dynamic stiffnesses were calculated from the slope of their l…
Effect of overground vs treadmill running on plantar pressure: Influence of fatigue
2013
Abstract The differences produced when running on a treadmill vs overground may call into question the use and validity of the treadmill as a piece of equipment commonly used in research, training, and rehabilitation. The aim of the present study was to analyze under pre/post fatigue conditions the effect of treadmill vs overground on plantar pressures. Twenty-seven recreational runners (17 men and 10 women) ran on a treadmill and overground at two speeds: S 1 = 3.33 m/s and S 2 = 4.00 m/s, before and after a fatigue protocol consisting of a 30-min run at 85% of their individual maximal aerobic speed (MAS). Contact time (CT in seconds), peak pressure (PP in kPa), and relative load (RL in …
