A human CCT5 gene mutation causing distal neuropathy impairs hexadecamer assembly in an archaeal model

Chaperonins mediate protein folding in a cavity formed by multisubunit rings. The human CCT has eight non-identical subunits and the His147Arg mutation in one subunit, CCT5, causes neuropathy. Knowledge is scarce on the impact of this and other mutations upon the chaperone's structure and functions. To make progress, experimental models must be developed. We used an archaeal mutant homolog and demonstrated that the His147Arg mutant has impaired oligomeric assembly, ATPase activity, and defective protein homeostasis functions. Performance Evaluation of CANIT Algorithm in Presence of Congestion Losses

In this paper, we analize by queuing-simulation CANIT (Congestion Avoidance with Normalized Interval of Time) algorithm performances in presence of congestion losses. In a former work [3], we proposed the algorithm (CANIT) for TCP (Transmission Control Protocol) congestion avoidance phase in order to improve fairness during this phase, and we showed that using CANIT algorithm in an environment without loss, instead of standard congestion avoidance algorithm improves both congestion avoidance fairness and bandwith utilization for long RTT connections. Implementing efficient TCP for the Internet necessarily has to cope with the problem that the source does not know in advance which window allocation policy should be the best to use for a given network condition. In this paper, an on-line adaptive fuzzy system is used at the source in an objective to find the best possible weighted combination among the available policies.

Biomonitoring exposure assessment to contemporary pesticides in a school children population of Spain

The exposure to pesticides amongst school-aged children (6-11 years old) was assessed in this study. One hundred twenty-five volunteer children were selected from two public schools located in an agricultural and in an urban area of Valencia Region, Spain. Twenty pesticide metabolites were analyzed in children's urine as biomarkers of exposure to organophosphate (OP) insecticides, synthetic pyrethroid insecticides, and herbicides. These data were combined with a survey to evaluate the main predictors of pesticide exposure in the children's population. It is expected that microvascular blood flow might be affected by blood glucose, blood insulin and C-peptide levels. In our investigation skin microvascular blood flow (LDF) was measured using laser doppler fluxometry at skin temperatures of 37 degrees C and 44 degrees C during a 75 g oral glucose load (OGT) or water in ten healthy volunteers (6 male, 4 female, age: 28.1+/-4.0) who had fasted overnight. Abstract In this paper, an automatic method for the screening of water samples containing Cu(II) was proposed, based on peryoxalate chemiluminescence reaction using coproporphyrin I as fluorophor compound to provide selectivity and a simple flow injection (FI) chemiluminescence detector (CLD). FI system conditions were chosen in order to distinguish samples over or under legislation limit established (50 μg l−1) with high reliability. The detection limit found was 9 μg l−1 and the linear dynamic range was 15–125 μg l−1 of Cu(II). Repeatibility and reproducibility studies gave good precision and accuracy with recovery near 100%. Abstract Dansylation of amines (primaries and secondaries) using C18 solid-phase supports is described. The time of analysis and the handing sample have been markedly improved with respect to those required for solution dansylation. The dansylated amine derivates were injected onto the liquid chromatography system and fluorescence detected. Arabidopsis TCP Transcription Factors Interact with the SUMO Conjugating Machinery in Nuclear Foci

In Arabidopsis more than 400 proteins have been identified as SUMO targets, both in vivo and in vitro. Among others, transcription factors (TFs) are common targets for SUMO conjugation. Here we aimed to exhaustively screen for TFs that interact with the SUMO machinery using an arrayed yeast two-hybrid library containing more than 1,100 TFs. We identified 76 interactors that foremost interact with the SUMO conjugation enzyme SCE1 and/or the SUMO E3 ligase SIZ1. These interactors belong to various TF families, which control a wide range of processes in plant development and stress signaling.
