Chromatin-dependent regulation of RNA polymerases II and III activity throughout the transcription cycle

The particular behaviour of eukaryotic RNA polymerases along different gene regions and amongst distinct gene functional groups is not totally understood. To cast light onto the alternative active or backtracking states of RNA polymerase II, we have quantitatively mapped active RNA polymerases at a high resolution following a new biotin-based genomic run-on (BioGRO) technique. Compared with conventional profiling with chromatin immunoprecipitation, the analysis of the BioGRO profiles in Saccharomyces cerevisiae shows that RNA polymerase II has unique activity profiles at both gene ends, which are highly dependent on positioned nucleosomes. Transcriptional Mechanisms of Proneural Factors and REST in Regulating Neuronal Reprogramming of Astrocytes

Summary Direct lineage reprogramming induces dramatic shifts in cellular identity, employing poorly understood mechanisms. Recently, we demonstrated that expression of Neurog2 or Ascl1 in postnatal mouse astrocytes generates glutamatergic or GABAergic neurons. Here, we take advantage of this model to study dynamics of neuronal cell fate acquisition at the transcriptional level. We found that Neurog2 and Ascl1 rapidly elicited distinct neurogenic programs with only a small subset of shared target genes. AbstractDiversification at the transcriptome 3’end is an important and evolutionarily conserved layer of gene regulation associated with differentiation and dedifferentiation processes. However the underlying mechanisms and functional consequences are poorly defined. The sex determining region of Chironomus thummi is associated with highly repetitive DNA and transposable elements.

The dominant male sex determiner in chromosome III of the midge Chironomus thummi thummi is closely linked to a large cluster of tandem-repetitive DNA elements, the Cla elements, which are otherwise highly repetitive and distributed over more than 200 sites on all chromosomes. Chromosome III displays a hemizygous cluster of Cla elements in males but not in females. The chromosomal location of this hemizygous Cla element cluster is in the region of the male determiner M as localized by cytogenetic analysis. Abstract Three different sampling units in current use and different sampling strategies were tested. Randomly placed microscope fields are good in estimating the daily mean concentration, but very big sample size is needed. Traverses across the slide in systematic order are best to estimate the shortterm concentrations and diurnal variation. A formula for the estimation of the error in one transverse traverse is given. Twelve transverse traverses in systematic order is also enough to estimate the daily mean concentration. Abstract The aim of this study was to evaluate the effects of a personal goal-based intervention on school and football self-determination motivation and satisfaction of newcomers within football training centres. Twenty-three trainees were divided randomly into two groups, either a treatment group or a control group. The treatment proposed to the participants was the Personal Goal Management Program. Trainees’ school and football motivation and satisfaction were measured before, during and after the intervention.
