Exploring organ-specific features of fibrogenesis using murine precision-cut tissue slices
2019
Fibrosis is the hallmark of pathologic tissue remodelling in most chronic diseases. Despite advances in our understanding of the mechanisms of fibrosis, it remains uncured. Fibrogenic processes share conserved core cellular and molecular pathways across organs. In this study, we aimed to elucidate shared and organ-specific features of fibrosis using murine precision-cut tissue slices (PCTS) prepared from small intestine, liver and kidneys. PCTS displayed substantial differences in their baseline gene expression profiles: 70% of the extracellular matrix (ECM)-related genes were differentially expressed across the organs. Culture for 48 h induced significant changes in ECM regulation and trig…
Colorectal cancer-derived small extracellular vesicles induce TGFβ1-mediated epithelial to mesenchymal transition of normal hepatocytes: new insights…
2023
More than 50% of patients affected by colorectal cancer (CRC) present liver metastasis, which is the most frequent cause of CRC-associated death. Numerous studies have shown that metastatic cascade is the result of complex mechanisms based on two-way interactions between invasive CRC cells and liver resident cells. In recent years, several findings have demonstrated that small extracellular vesicles (SEVs) released by cancer cells play a crucial role in the formation of pre-metastatic niche in the liver, specifically affecting the activities of non-parenchymal cells as Kupffer cells and hepatic stellate cells. However, although hepatocytes (heps) are the most conspicuous in the liver, their…
Mboat7 down-regulation by hyper-insulinemia induces fat accumulation in hepatocytes.
2020
Background: Naturally occurring variation in Membrane-bound O-acyltransferase domain-containing 7 (MBOAT7), encoding for an enzyme involved in phosphatidylinositol acyl-chain remodelling, has been associated with fatty liver and hepatic disorders. Here, we examined the relationship between hepatic Mboat7 down-regulation and fat accumulation. Methods: Hepatic MBOAT7 expression was surveyed in 119 obese individuals and in experimental models. MBOAT7 was acutely silenced by antisense oligonucleotides in C57Bl/6 mice, and by CRISPR/Cas9 in HepG2 hepatocytes. Findings: In obese individuals, hepatic MBOAT7 mRNA decreased from normal liver to steatohepatitis, independently of diabetes, inflammatio…
Susceptibility to collagen-induced arthritis is modulated by TGFβ responsiveness of T cells
2004
The objective of our study was to determine the regulatory effects that endogenous transforming growth factor beta (TGFbeta) exerts on T cells in the pathogenesis of collagen-induced arthritis (CIA). CIA was induced in transgenic mice expressing a dominant negative TGFbeta type II receptor in T cells under the control of the human CD2 promoter. Clinical and histological arthritis scores were determined and experiments on disease induction and the healing phase of disease were performed. The proliferation and cytokine production of draining lymph node cells in vitro were analyzed. Transgenic mice were more susceptible to induction of CIA. The overall incidence was higher in transgenic mice t…

Ustekinumab therapy changes the transcriptional activity pattern of TGF-β1–3 genes
2019
Introduction One of the examples of genes whose expression can be altered by the action of ustekinumab is TGF-β. It is a pleiotropic cytokine whose activity affects psoriatic changes and the state of homeostasis of the whole organism. Aim To evaluate the effect of ustekinumab on the transcriptional activity of TGF-b family genes in patients with psoriatic arthritis and to check whether the results obtained can be helpful in monitoring the progress of treatment. Material and methods From total PBMCs obtained from peripheral blood of 14 patients with psoriatic arthritis, total RNA was isolated. The expression level of the TGF-β1, TGF-β2 and TGF-β3 genes was determined by RT-qPCR in real time.…
