A rapid and cost-efficient DMSO-based method for isolating DNA from cultured lichen photobionts

We have developed a simple and fast procedure for the purification of PCR-quality DNA from cultured lichen photobionts. This new one-step method uses the solvent dimethyl sulfoxide (DMSO) combined with heat treatment to chemically breakdown algal and plant tissues. The DMSO-DNA extracts may be directly precipitated and purified by standard techniques in a total time of approximately 30 min. Compared to other DNA extraction protocols, the DMSO-based method suppresses the need for liquid nitrogen, any extraction buffer, grinding, phenol, and long incubation or centrifugation steps, thereby considerably reducing the possibility of contamination. A novel restricted access cation exchanger with sulphonic acid groups at the internal surface was proven to be highly suitable in the sample clean up of peptides on-line coupled to HPLC-electrospray ionization (ESI)-MS. Neuropeptide Y (NPY) and several of its fragments in plasma were subjected to the sample clean-up procedure. Isobaric vapor−liquid equilibria for the binary system chloroform + methanol as well as the vapor−liquid equilibria for the chloroform + methanol + [emim][triflate] ternary system have been obtained at 100 kPa using a recirculating still. The ternary ionic liquid (IL) system presents an immiscibility zone at high chloroform composition, which increases with the temperature. Protoplasts fromPodospora anserina: Isolation, purification, and transformation

Protoplasts fromPodospora anserina mycelium were produced using the commercially available enzyme Novozym 234. Different parameters involved in protoplast isolation were analyzed in order to establish optimal conditions, and protoplast production was notably increased. For the purification of protoplasts, several techniques based on both centrifugation and filtration were assayed, with filtration yielding the best results. Regeneration of protoplasts was studied on different media and osmostic stabilizers, and about 80% regeneration was obtained. Pauson-Khand reaction of internal dissymmetric trifluoromethyl alkynes. Influence of the alkene on the regioselectivity.

Abstract: The scope of the Pauson-Khand reaction (PKR) of internal trifluoromethyl alkynes, previously described with norbornadiene, is expanded to norbornene and ethylene. A thorough structural analysis of the resulting PK adducts has been carried out to unveil that α-trifluoromethylcyclopentenones are preferred in all cases, independently of the electronic properties of the alkyne. The regioselectivity observed with norbornadiene and ethylene is higher than in the case of norbornene.

Oligopeptides with the sequence Z(GlyProPro)nOBut (n = 3–7) were synthesized along a clasical pathway. Generally long fragments were condensed in order to allow an easy separation of the products from the reactants. It is established by molecular weight determination and measurements of circular dichroism that the peptides form the collagen-like triple helix in methanol and that they assume the random coil conformation in dilute acetic acid. The circular dichroism spectra agree reasonably well with the corresponding spectra for (ProProGly)10 which was obtained by solid phase synthesis in another laboratory.

Existence of metastable intermediate lysozyme conformation highlights the role of alcohols in altering protein stability.

Alcohols have a manifold effect on the conformational and thermodynamic stability of native proteins. Here, we study the effect of moderate concentrations of trifluoroethanol (TFE) on the thermal stability of hen egg-white lysozyme (HEWL), by far-UV circular dichroism and by steady-state and time-resolved photoluminescence of intrinsic tryptophans. Our results highlight that TFE affects lysozyme stability by preferential solvation of the protein molecule. Furthermore, we discovered the existence at 20% TFE of an equilibrium partially folded state of lysozyme, intermediate between the native and the unfolded state. Environmental pollution with chiral organic compounds is an emerging problem requiring innovative sensing methods. Amino-functionalized thioureas, such as 2-(dimethylamino)cyclohexyl-(3,5-bis(trifluoromethyl)phenyl)thiourea (Takemoto’s catalyst), are widely used organocatalysts with virtually unknown environmental safety data. Ecotoxicity studies based on the Vibrio fischeri luminescence inhibition test reveal significant toxicity of Takemoto’s catalyst (EC50 = 7.9 mg/L) and its NH2-substituted analog (EC50 = 7.2–7.4 mg/L). Isolation and characterization of haemoporin, an abundant haemolymph protein from Aplysia californica.

In the present study, we show the isolation and characterization of the protein haemoporin, which constitutes the second most abundant protein fraction in the haemolymph of the marine gastropod Aplysia californica. Although Aplysia is commonly used to investigate the molecular basis of learning, not much is known about the proteins in its haemolymph, which is in contact with the neurons owing to the open circulatory system of molluscs.
