An Optimized and Rapid DNA Extraction Method From Leaves of Grapevine Suitable for PCR-DGGE Based Analysis
2010
Molecular techniques and genetic studies require a fair amount of DNA of high quality in order to produce reliable and clear results (Li et al., 2007). In grapevine, the yield and quality of DNA can be significantly affected by secondary metabolites such as polyphenols, polysaccharides and tannins, that may be consistently abundant during the different stages of leaf development (Iandolino et al., 2004). The development of a simple, rapid and reliable method for the extraction of genomic DNA from grape leaves collected at several stages during development, as well as from healthy grape leaves and leaves infected by pathogenic microorganisms, was the main objective of this study. The protoco…
Vascular flora evolution in the Soqotra Archipelago (Indian Ocean)
2012
The main floristic and vegetational features of the Soqotra Archipelago are outlined. The theories of vicariance and dispersal are commented with the support of examples suggesting the idea that both are complementary in the establishment and evolution of the flora of Soqotra. Finally the relation of alien vs natural elements of the flora is analyzed.
Ellenberg Indicator Values for the vascular flora alien to Italy
2018
Studies to date about plants alien to Italy have had limited focus on the ecology of this component of the flora. Ellenberg's indicator values are a useful tool to delineate the relationship between plants and environment, recognizing for each species a functional role as biological indicator; these values have been proposed for estimating the influence of the main environmental factors in determining flora and vegetation changes on a specific surface area. This contribution includes a list of 1206 taxa of plants naturalized in at least one administrative region or casual in at least three regions of Italy. In addition, some methodological considerations on the attribution and use of Ellenb…

Batch Test Evaluation of Four Organic Substrates Suitable for Biological Groundwater Denitrification
2014
Nitrates pollution represents nowadays a serious issue related to the quality of groundwater; continuous growth of industrial-scale agricultures lead to an increase of nitrates content in groundwater in the last years. Several technologies have been validated as capable to promote in situ biological nitrates remediation, such as permeable reactive barriers (PRB), biotrench, biobarriers etc. These technologies are all characterised by the use of organic substrate that act as a slow release carbon source. In free dissolved oxygen absence, such organic carbon is further oxidised, by heterotrophic bacteria naturally present in soil, in compliance to anoxic metabolism by using nitrates bound oxy…
Mathematical Modelling of In-sewer Processes as a Tool for Sewer System Design
2018
The objective of this paper is to evaluate the potential impact of in-sewer processes (COD components transformation and hydrogen sulphide production) on the design of sewer systems. The tool used for such analysis is a mathematical model derived from the WATS model (Wastewater Aerobic/anaerobic Transformation in Sewers) able to describe the processes occurring in the sewer system both under aerobic and anaerobic conditions. The model is applied to three synthetic catchments with, respectively, 10,000, 50,000 and 250,000 inhabitants connected to gravity sewer systems different in terms of type (separate or combined), slope, length, travel time, wastewater temperature. The simulation results…
Phytoplankton distribution along trophic gradients within and among reservoirs in Catalonia (Spain)
2008
SUMMARY 1. Longitudinal gradients in the epilimnetic waters of stratified reservoirs provide a useful database to study changing environmental conditions. The spatial distribution, assemblage structure and specific adaptations of phytoplankton assemblages can be analysed along these gradients over short time scales. 2. Four reservoirs with a similar typology, located along an altitudinal gradient in the same eco-region, were sampled along their longitudinal axes. In total, 19 sampling stations provided a trophic spectrum, ranging from oligo-mesotrophy to hypertrophy, which was quantified by calculating the trophic state index of each sampling station in the four reservoirs. 3. Several patte…
Gestione sostenibile delle foreste Mediterranee e uso energetico delle biomasse forestali residuali
2014
he book describes the reasons that led the Regional Department of Rural and Territorial Development to take part in the PROFORBIOMED Project. They can be summarized by the need to dispose of a tool for the sustainable management of all the state-owned forests of the Region. As a matter of fact, the Project aims at developing a model of sustainable forest management, through the recovery and reuse of wood scraps from ordinary silvicultural operations, to be used for the production of power and heat inside of a process adopting natural renewable energy sources. The main actions taken and the methodologies adopted are described, as well as the principles and instruments required for the settin…
Artificial chromosomes for antibiotic-producing actinomycetes.
2000
Bacteria belonging to the order Actinomycetales produce most microbial metabolites thus far described, several of which have found applications in medicine and agriculture. However, most strains were discovered by their ability to produce a given molecule and are, therefore, poorly characterized physiologically and genetically. Thus, methodologies for genetic manipulation of actinomycetes are not available and efficient tools have been developed for just a few strains. This constitutes a serious limitation to applying molecular genetics approaches to strain development and structural manipulation of microbial metabolites. To overcome this hurdle, we have developed bacterial artificial chrom…
