Proteasome inhibitors, like MG132, can exert cell growth inhibitory and apoptotic effects in different tumor types. The immunonegative staining procedure has been utilized for the study of epitope localization on immune complexes formed from keyhole limpet hemocyanin type 2 (KLH2) di- and multidecamers, and the 20S and 26S proteasome from Xenopus laevis. Complex Regional Pain Syndrome (CRPS) occurs in about 2% of patients after fracture of the limbs. In an earlier clinical study with 102 probands we have shown that the serum protease network in CRPS might be less effective. Based on these results we hypothesized that angiotensin-converting enzyme (ACE) and carboxypeptidase N (CPN) activity contribute to the differences of labeled bradykinin (DBK) degradation by patients' sera. Details of the enzymatic processes remained however unclear. The contributions of ACE and CPN in the serum degradation of DBK were studied using specific inhibitors. Proteolytic capacity in mouse cardiac muscle was analyzed 1, 3, and 6 days after exhaustive intermittent or submaximal prolonged running. No significant changes were recorded in the activities of acid or alkaline proteases, β-glucuronidase or trypsin inhibitor. Similarly, no changes were found in the rates of acid or neutral autolysis.
