Lamotrigine use in pregnancy and risk of orofacial cleft and other congenital anomalies

Objective: To test previous signals of a risk of orofacial cleft (OC) and clubfoot with exposure to the antiepileptic lamotrigine, and to investigate risk of other congenital anomalies (CA).Methods: This was a population-based case–malformed control study based on 21 EUROCAT CA registries covering 10.1 million births (1995–2011), including births to 2005 in which the clubfoot signal was generated and a subsequent independent study population of 6.3 million births. A total of 226,806 babies with CA included livebirths, stillbirths, and terminations of pregnancy following prenatal diagnosis. Short X···N Halogen Bonds With Hexamethylenetetraamine as the Acceptor

Hexamethylenetetramine (HMTA) and N-haloimides form two types of short (imide)X···N and X–X···N (X = Br, I) halogen bonds. Nucleophilic substitution or ligand-exchange reaction on the peripheral X of X–X···N with the chloride of N-chlorosuccinimide lead to Cl–X···N halogen-bonded complexes. The 1:1 complexation of HMTA and ICl manifests the shortest I···N halogen bond [2.272(5) Å] yet reported for an HMTA acceptor. Two halogen-bonded organic frameworks are prepared using 1:4 molar ratio of HMTA and N-bromosuccinimide, each with a distinct channel shape, one possessing oval and the other square grid. Conformational properties of cyanomethoxy calix[4]arenes.

O-Alkylation of the dinitro calix[4]arene 2, easily available by selective ipso-nitration of the di-cyanomethyl ether 1, with allylbromide (DMF/Cs2CO3) gave tetraethers 3 and 4 with anti- and syn-orientations of the two allyl ether residues. The two possible stereoisomers of 3 in the partial cone and 1,2-alternate conformation exist as an equilibrium mixture which could be quantitatively analysed by 1H NMR spectroscopy. The temperature dependence of this equilibrium leads to ΔH0 = − 7.6 to −9.7 kJ mol−1 in different solvents (tetrachloroethane, benzene, dimethylsulfoxide).
