The effects of oxotremorine and atropine on the release of acetylcholine from longitudinal muscle strips of the guinea-pig ileum stimulated at frequencies between 0.1 and 3 Hz in the presence of eserine were investigated. In control experiments the acetylcholine output per stimulus declined with increasing frequencies of stimulation. 2. Oxotremorine inhibited the release of acetylcholine in a concentration-dependent fashion. At a concentration of 10−6 M oxotremorine, the release evoked by 0.1 Hz was reduced by 54%. Effects of Different Glucose Concentrations on Spinal Anaesthesia with Bupivacaine and Tetracaine

The effects of 5% and 8% glucose in 0.5% tetracaine or bupivacaine on the anaesthetic spread were investigated in 80 urological patients requiring spinal anaesthesia for trans-urethral resection of the prostate. The local anaesthetic solutions were randomly administered, the patients being divided into four groups of 20, and the anaesthetic profile was then evaluated in a double-blind fashion by an independent observer. Maximum cephalad spread of analgesia was significantly greater with tetracaine in 8% glucose compared to the other three groups (tetracaine/5% glucose, bupivacaine/5 or 8% glucose) (P less than 0.05). Determination of procaine and tetracaine in plasma samples by micellar liquid chromatography and direct injection of sample

A liquid chromatographic procedure is proposed for the determination of procaine and tetracaine in plasma samples with direct injection. The method uses a Spherisorb octadecylsilane ODS-2 C18 analytical column and a micellar mobile phase containing 0.15 M sodium dodecyl sulphate, 0.5% triethylamine at pH 2.5 and 10% propanol. The UV detection was carried out at 300 nm. Plasma sample preparation required only adequate dilution with the mobile phase before injection into the chromatographic system. The proposed method allows the determination of procaine and tetracaine in plasma at therapeutic levels.

Effects of pentobarbital on acetylcholine (ACh) release, force of contraction and nervous conduction were studied in isolated heart preparations and in cervical vagus nerves, respectively. 4-Aminopyridine and tetracaine were used as pharmacological tools to eludicate the mode of action of pentobarbital. 1. 4-Aminopyridine (10−4 M) markedly increased the overflow of ACh from the isolated chicken heart evoked by electrical stimulation (1–50 Hz, 1 ms, 40 V) of the cervical vagus nerves. This effect of 4-aminopyridine was highest at low frequencies of stimulation (+ 226% at 1 Hz) and declined with increasing frequencies to reach a minimum augmentation of 22% at 30 Hz. 2. Effects of Propofol (Intravenous Propofol Emulsion) on Cell Membranes Measured by Electrofusion and Electroporation

The influence of propofol (CAS 2078-54-8 (intravenous propofol emulsion) on cell membrane properties was investigated in vitro with techniques of cell electrofusion and cell electroporation. Human lymphoma cells and plant protoplasts were chosen as a model system. Propofol (intravenous propofol emulsion) decreased the electrofusion yield of the cells and their membrane permeability. A 50% decrease in relative electrofusion was observed in human lymphoma cells in the presence of about 0.05 mmol/l propofol (intravenous propofol emulsion) and in plant protoplasts in the presence of about 0.1 mmol/l.
