Cyclodextrin‐Calixarene Nanosponges as Potential Platforms for pH‐Dependent Delivery of Tetracycline

Four mixed cyclodextrin-calixarene nanosponges were tested as possible Drug Delivery Systems, using Tetracycline antibiotic as a suitable model drug. The selected nanosponges featured a different composition ratio between the two host co-monomer components, and the possible presence of ionisable amine or carboxyl groups deriving from chemical post-modification. We have tested whether tetracyclines (TETs) are able to protect mice from lipopolysaccharide (LPS)-induced shock, a cytokine-mediated inflammatory reaction. Mice, injected with a single dose of tetracycline base (TETb; 1.5, 10 and 20 mg/kg of body weight) or doxycycline (DOXY; 1.5 mg/kg), were significantly protected from a lethal intraperitoneal injection of LPS (500 micrograms per mouse). TETs acted in early events triggered in response to LSP; in fact, they were no longer significantly protective if injected more than 1 h after the injection of endotoxin. Photolytic and TiO2-Assisted Photodegradation of Aqueous Solutions of Tetracycline

Aqueous solutions of tetracycline were UV-irradiated in homogeneous and heterogeneous systems. Tetracycline disappeared in both systems, but the presence of TiO2 accelerated the photodegradation of the antibiotic in comparison with direct photolysis. The rate of decomposition was dependent on the concentration of tetracycline and its degradation on the surface of TiO2 can be described by means of a pseudo-first order kinetics. Photolysis involved only the photodeamination of tetracycline and a partial mineralization. Unlikely, the treatment with TiO2 and UV-light resulted in a complete mineralization of the antibiotic drug.
