5-HT3 receptor-channels coupled with Na+ influx in human T cells: role in T cell activation.

The study was conducted on a human (Jurkat) T cell line, loaded with a Na+ fluorescent probe, SBFI/AM. Serotonin and an agonist of 5-HT3 receptor-channels, 2-methyl-5HT, evoked Na+ influx, whereas the agonists of other serotonergic receptor subtypes, i.e., 5-HT1A and 5-HT1B receptors, failed to induce Na+ influx in these cells. By using 3H-BRL43694, an agonist of 5-HT3 receptor-channels, we characterized 5-HT3 lymphocyte receptors which exhibited a density (Bmax) of 300 +/- 20 fmol/10(6) cells and a Kd of 30 nM in Jurkat T cells. Characterization of 5-Hydroxytryptamine receptors in goat cerebral arteries

In isolated goat middle cerebral artery segments, 5-hydroxytryptamine (5-HT, 10(-8)-3 x 10(-5) M) elicited concentration-dependent contractions with EC50 = 2.1 (1.9-2.5) x 10(-7) M and Emax = 64 +/- 2% of 50 mM KCl-induced contraction. 2. New findings show existence of FGFR1-5-HT1A heteroreceptor complexes in 5-HT nerve cells of the dorsal and median raphe nuclei of the rat midbrain and hippocampus. Synergistic receptor-receptor interactions in these receptor complexes indicated their enhancing role in hippocampal plasticity. The existence of FGFR1-5-HT1A heteroreceptor complexes also in midbrain raphe 5-HT nerve cells open up the possibility that antidepressant drugs by increasing extracellular 5-HT levels can cause an activation of the FGF-2/FGFR1 mechanism in these nerve cells as well. The ascending midbrain 5-HT neurons known to contain 5-HT1A autoreceptors may be dysregulated in depression due to a reduced trophic support. With in situ proximity ligation assay (PLA) and supported by co-location of the FGFR1 and 5-HT1A immunoreactivities in midbrain raphe 5-HT cells, evidence for the existence of FGFR1–5-HT1A heteroreceptor complexes were obtained in the dorsal and median raphe nuclei of the Sprague–Dawley rat. Their existence in the rat medullary raphe RN33B cell cultures was also established. Abstract Cracking of tetralin and decalin was carried out over several zeolites to establish the effect of the pore topology of the catalyst on product distribution. These molecules were chosen as probe molecules, because they indicate which catalyst is the best for cracking or hydrotreating the light cycle oil (LCO) fraction, which is obtained directly from fluid catalytic cracking units. Cu/P(4-PVP) stereochemistry in tetralin liquid-phase oxidation

Abstract Cu/P(4-PVP) catalyst was used as catalyst for oxidation of tetralin in liquid phase. The catalyst was prepared by reaction of a Cu(II) salt with a 4-vinylpyridine-co-divinylbenzene polymer receptor and characterized by solid 1 H and 13 C NMR, XPS, FTIR, ESR spectroscopies, and by chemical reaction with pure tetralin. That information was used to gain insight into the stereochemistry of the active site needed to orient the reaction toward tetralone production. It was found that the Cu coordination with the pyridine was critical in influencing the peroxy-radical catalytic reaction, and that polyvinylpyridine was able to limit the autothermal oxidation. The enantioselective synthesis of tetralin derivatives bearing a trifluoromethylated all‐carbon quaternary stereocenter has been accomplished through a synthetic sequence comprising an organocatalytic β‐functionalization of ortho‐1‐trifluoromethylvinyl‐(hetero)aromatic conjugated aldehydes followed by the intramolecular nitrone 1,3‐cycloaddition reaction (INCR). Both nitromethane and N‐Cbz‐hydroxylamine were employed as nucleophiles in the initial organocatalytic conjugate addition step, which provided the chiral information required for the subsequent diastereoselective INCR.
