Stabilization of an enzymatic extract from Penicillium camemberti containing lipoxygenase and hydroperoxide lyase activities

International audience; The stabilization of an enzymatic extract, obtained from Penicillium camemberti containing lipoxygenase (LOX) and hydroperoxide lyase (HPL) activities, was investigated using selected additives. Although the addition of KCl (86%, w/w) to the enzymatic extract decreased slightly (7%) the LOX activity, it increased HPL activity by 2.25 fold; however, the addition of dextran resulted in the inactivation of both enzymatic activities. The stability of the solid lyophilized enzymatic extract was greater in the presence of KCl than that without it, with ∼100% residual activity after 8 and 4 weeks of storage at −80 °C, for LOX and HPL, respectively. Abstract The structure of the surface layer of materials is usually quite different from the bulk. Detailed information about the surface structure is often not available. This, together with the complex nature of low temperature plasma treatment, hinders the investigations of plasma chemical processes on solid surfaces. Monomolecular Langmuir-Blodgett (LB) structures offer a unique opportunity to prepare model surfaces with known thickness and molecular architecture. Porous materials as delivery and protective agents for Vitamin A

The suitability of porous materials to immobilize and release under control bioactive molecules prompted us to design and study delivery systems of Vitamin A (VitA). This molecule, relevant in several physiological functions, is easily oxidized. Commercial VitA was immobilized in two different clays, montmorillonite K-10 (MMT) and sepiolite (SEP), and in MCM-41, by impregnation. Characterization of the resulting hybrid materials by XRD, FTIR and 13C and 29Si (MAS) NMR spectroscopies revealed its presence. AbstractThree isoforms of the major light-harvesting chlorophyll (Chl) a/b complexs of photosystem II (LHCIIb) in the pea, namely, Lhcb1, Lhcb2, and Lhcb3, were obtained by overexpression of apoprotein in Escherichia coli and by successfully refolding these isoforms with thylakoid pigments in vitro. The sequences of the protein, pigment stoichiometries, spectroscopic characteristics, thermo- and photostabilities of different isoforms were analysed. Mutational robustness has important evolutionary implications, yet the mechanisms leading to its emergence remain poorly understood. One possibility is selection acting on a correlated trait, as for instance thermostability (plastogenetic congruence). Here, we examine the correlation between mutational robustness and thermostability in experimental populations of the RNA bacteriophage Qβ. Thermostable viruses evolved after only six serial passages in the presence of heat shocks, and genome sequencing suggested that thermostability can be conferred by several alternative mutations. Mutations enhancing the thermostability of β-glucosidase A of Bacillus polymyxa, a family 1 glycosyl hydrolase, have been obtained after hydroxylamine mutagenesis of a plasmid containing the bglA gene, transformation of Escherichia coli with the mutagenized plasmid, and identification of transformant colonies that showed β-glucosidase activity after a thermal treatment that inactivated the wild-type enzyme. Two additive mutations have been characterized that cause replacement of glutamate at position 96 by lysine and of methionine at position 416 by isoleucine respectively. The chicken avidin gene family consists of avidin and seven separate avidin-related genes (AVRs) 1-7. Avidin protein is a widely used biochemical tool, whereas the other family members have only recently been produced as recombinant proteins and characterized. In our previous study, AVR4 was found to be the most stable biotin binding protein thus far characterized (T(m) = 106.4 degrees C). In this study, we studied further the biotin-binding properties of AVR4. AbstractInformation on protein internal motions is usually obtained through the analysis of atomic mean-square displacements, which are a measure of variability of the atomic positions distribution functions. We report a statistical approach to analyze molecular dynamics data on these displacements that is based on probability distribution functions. Using a technique inspired by the analysis of variance, we compute unbiased, reliable mean-square displacements of the atoms and analyze them statistically.
