Bacterial community analysis of a gas-phase biotrickling filter for biogas mimics desulfurization through the rRNA approach

Abstract The bacterial composition of a lab-scale biotrickling filter (BTF) treating high loads of H 2 S was investigated by the rRNA approach. Two 16S rRNA gene clone libraries were established 42 and 189 d after reactor startup, while fluorescent in-situ hybridization (FISH) with DNA probes was performed throughout 260 d of reactor operation. Diversity, community structure and metamorphosis were studied from reactor startup to fully-established pseudo-steady state operation at near neutral pH and at an inlet H 2 S concentration of 2000 ppmv (load of 55.6 g H 2 S m −3 h −1 ). Local dynamics and solute-solvent exchange properties of rusticyanin (Rc) from Thiobacillus ferrooxidans have been studied by applying heteronuclear ((1)H, (15)N) NMR spectroscopy. (15)N relaxation parameters have been determined for the reduced protein, and a model-free analysis has been applied. The high average value of the generalized order parameter, S(2) (0.93), indicates that Rc is very rigid. The analysis of cross correlation rates recorded in both the reduced and the oxidized forms conclusively proves that Rc possesses the same dynamic features in both oxidation states. This review deals with different aspects concerning the genus Thiobacillus as an obligat chemoautotroph able to oxidize reduced anorganic sulphur compounds. There are given the following topics: physiology of the genus (nutritional requirements, enzymatic complexes for sulphur oxidation, energy generation, reductive CO2 assimilation and growing in presence of organic compounds). The ecological significance of Thiobacilli in the sulphur cycle in Nature is presented.
