Bioaccumulation and subchronic physiological effects of waterborne iron overload on whitefish exposed in humic and nonhumic water.

One-year-old whitefish, Coregonus lavaretus, were exposed to three types of iron-rich water, two dilutions for each, in a subchronic (30-day) experiment. In natural iron-rich humic water, both the bioaccumulation and physiological effects of iron exposure were negligible. In humic-free water with high amount of additional inorganic iron (nominally 8 mg Fe/L), Fe accumulated in gills, liver, and gut. This accumulation was accompanied by decreased glycogen phosphorylase activities and microsomal EROD activity in the liver as well as decreased plasma sodium and potassium concentrations. Abstract European eels ( Anguilla anguilla ) were exposed to a sublethal thiobencarb concentration of 0.22 mg/L in a flow-through system for 96 h. Mg 2+ and Na + –K + adenosine triphosphatase (ATPase) activities were evaluated in gill and muscle tissues at 2, 12, 24, 48, 72, and 96 h of thiobencarb exposure. Gill ATPase activities were rapidly inhibited from 2 h of contact onward. Highest inhibition was registered for Na + , K + -ATPase (85%) from 2 to 12 h. Both Mg 2+ and total ATPase were inhibited (>73%) during the first hours of toxicant exposure. [EN] Mangiferin, a natural compound isolated from Mangifera indica L, was incorporated in glycerosomes, ethosomes and alternatively in glycerol-ethanol phospholipid vesicles (glycethosomes). Actually, only glycethosomes were able to stably incorporate the mangiferin that was loaded at increasing concentrations (2, 4, 6, 8 mg/mL). The morphology, size distribution, rheological properties, surface charge and entrapment efficiency of prepared vesicles were deeply measured. Rifampicin loaded glycerosomes, vesicles composed of phospholipids, glycerol and water, were combined with trimethyl chitosan chloride (TMC) to prepare TMC-glycerosomes or, alternatively, with sodium hyaluronate (HY) to obtain HY-glycerosomes. These new hybrid nanovesicles were tested as carriers for pulmonary delivery of rifampicin. Glycerosomes without polymers were also prepared and characterized. All vesicles were similar: they were spherical, multilamellar and able to incorporate good amount of rifampicin (EE%∼55%). The binding and bioavailability of pyrene was studied in the laboratory in two humic fresh waters and in a reference water without dissolved organic material (DOM), measured as dissolved organic carbon (DOC). The uptake of pyrene by Daphnia magna in short-term (24 h) accumulation experiments was fitted to a first-order rate-kinetic equation to calculate simultaneous uptake and elimination rates. The partition coefficients of pyrene to DOC (KDOC) were 37.1 x 103 in Pielisjoki River (9.4 mg DOC L(-1)), and 34.9 x 103 in Lake Kontiolampi (17.4 mg DOC L(-1)) waters, indicating similar binding affinities of pyrene for both humic waters. This work reports significant advances on the transport in hepatic cells of resveratrol, a natural polyphenol with potent protective properties. First, we describe a new simple technique to qualitatively follow resveratrol cell uptake and intracellular distribution, based on resveratrol fluorescent properties. Second, the time-course study and the quantification of (3)H-labelled resveratrol uptake have been performed using human hepatic derived cells (HepG2 tumor cells) and hepatocytes. To provide a basis for the investigation of the intermediate filament (IF) protein vimentin in one of the most promising experimental vertebrate systems, the zebrafish (Danio rerio), we have isolated a cDNA clone of high sequence identity to and with the characteristic features of human vimentin. Using this clone we produced recombinant zebrafish vimentin and studied its assembly behaviour. Unlike other vimentins, zebrafish vimentin formed unusually thick filaments when assembled at temperatures below 21 degrees C.
