Transfer of plasmid DNA into cells with microelectroporation arrays on a chip

2000

The possibility to transfer pure DNA into bacterial cells forms the basis for the genetic engineering of the cell. Electroporation is a powerful and easy technique to introduce plasmid DNA into cells. Its drawback for use with high-throughput approaches is that with standard electroporation chambers the reactions have to carried out one after the other and the electroporation cuvettes are expensive. To obtain the possibility of high-throughput electroporation reactions Escherichia coli cells were electroinjected in parallel with different plasmids in reactions as small as 100 nl on a microstructured array of electrodes, forming hundred separate electroporation units on a chip of a square in…

Occurrence of rhizobia in the gut of the higher termite Nasutitermes nigriceps

2006

Wood-eating termites feed on a diet highly deficient in nitrogen. They must complement their diet with the aid of nitrogen-fixing bacteria. Nitrogen fixation in the gut has been demonstrated, but information about nitrogen-fixing bacteria in pure culture is scarce. From the higher termite Nasutitermes nigriceps the symbiotic bacterial strain M3A was isolated, which thrives in the hindgut contents. The Gram-negative strain exhibited similarities to the species of the genus Ensifer (including Sinorhizobium) on the basis of morphological and physiological/biochemical features. The 16S rRNA gene analysis showed the highest sequence similarity of the isolate M3A to Ensifer adhaerens (>99%; ATCC …

Maximal ℓ p ‐regularity of multiterm fractional equations with delay

2020

[EN] We provide a characterization for the existence and uniqueness of solutions in the space of vector-valued sequences l(p) (Z, X)for the multiterm fractional delayed model in the form Delta(alpha)u(n) + lambda Delta(beta)u(n) = Lambda u(n) + u(n-tau) + f(n), n is an element of Z, alpha, beta is an element of R+, tau is an element of Z, lambda is an element of R, where X is a Banach space, A is a closed linear operator with domain D(A) defined on X, f is an element of l(p)(Z,X) and Delta(Gamma) denotes the Grunwald-Letkinov fractional derivative of order Gamma > 0. We also give some conditions to ensure the existence of solutions when adding nonlinearities. Finally, we illustrate our resu…

Foxp3 Silencing with Antisense Oligonucleotide Improves Immunogenicity of an Adjuvanted Recombinant Vaccine against Sporothrix schenckii

2021

Made available in DSpace on 2021-06-25T10:56:14Z (GMT). No. of bitstreams: 0 Previous issue date: 2021-04-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Background: In recent years, there has been great interest in developing molecular adjuvants based on antisense oligonucleotides (ASOs) targeting immunosuppressor pathways with inhibitory effects on regulatory T cells (Tregs) to improve immunogenicity and vaccine efficacy. We aim to evaluate the immunostimulating effect of 2′OMe phosphorothioated Foxp3-targeted ASO in an antifungal adjuvanted recombinant vaccine. Methods: The uptake kinetics of Foxp3 ASO, its cyto-toxicity and its ability to deplete Tregs were evaluated in…

Antiviral activity of aged green tea extract in model food systems and under gastric conditions.

2018

Aged-green tea extract (GTE) is known to reduce the infectivity of hepatitis A virus (HAV) and murine norovirus (MNV), a human norovirus surrogate, in vitro and in washing solutions. Initially, the effect of aged-GTE was evaluated on virus like particles (VLPs) of human norovirus (HuNoV) genogroup I (GI) by a porcine gastric mucine (PGM)-enzyme-linked immunosorbent assay (ELISA) and transmission electron microscopy (TEM), and on HuNoV GI suspensions by an in situ capture-RT-qPCR method, suggesting that HuNoVs are very sensitive to aged-GTE treatment at 37 °C. Moreover, the potential application of aged-GTE was evaluated using model foods and simulated gastric conditions. Then, aged-GTE samp…
