Abstract Phylogenetic relationships of the African toad Bufo danielae are investigated using a variety of biochemical approaches. Nuclear DNA content was assayed and compared to representatives of three species groups of African Bufo. Cellogel electrophoresis of plasma proteins was performed and patterns of B. danielae compared with those of representatives of the African B. regularis species complex. Finally microcomplement fixation analyses of albumin relationships of B. danielae and African Bufo were carried out. The strengths of the varied approaches for phylogenetic analysis are discussed. Quantitative behavioral experiments have shown that the toad uses mainly two types of gestalt information in prey/enemy discrimination: pattern extension in the direction of movement promotes, in general, the signal value prey, while extension perpendicular to the direction of movement promotes that of enemy. Summary The β-ketoadipate pathway is the major route for degradation of aromatic compounds by various soil microorganisms. Protocatechuate 3,4-dioxygenase, a key enzyme of this pathway and which is encoded by pcaGH genes, catalyses the ring cleavage of protocatechuate. Microorganisms harbouring pcaGH genes are widely distributed in the environment but little is known about their relative abundance within the total microflora. Hence, this paper reports the development of a real-time PCR assay to quantify the bacterial pcaH sequence that encodes the β sub-unit of the protocatechuate 3,4-dioxygenase. The population of the common toad, Bufo bufo, on Jersey (Channel Islands, UK) has undergone a marked decline over recent years. Hypothesized causes for this decline include land use change, pollution, climate change and predation. We examined the carcasses of 97 wild adult common toads for gastrointestinal and pulmonary helminths. In addition, 31 adult toad carcasses were examined for the presence of ranavirus infection, and 212 wild common toads were examined for the presence of infection with the fungus Batrachochytrium dendrobatidis.
