Introduction: Diffusion tensor imaging (DTI) is the most commonly used technique to extract microstructural features from a set of diffusionweighted images. In addition to the metrics obtained with DTI, diffusion kurtosis imaging (DKI) can provide non-Gaussian diffusion measures by means of the kurtosis tensor. The Hermann’s tortoise is a strictly European species found mostly in areas with Mediterranean and sub-Mediterranean climes and, especially the subspecies T. hermanni hermanni, has to deal with various threats which reduced its geographic distribution to only a few isolated populations. This paper explores the presence of tortoise remains and its meaning in Palaeolithic archaeological contexts. We focus on two topics: (1) the presence or absence of the genus Testudo from the Early Pleistocene to the Holocene in SW Europe; and (2) the relationship between Testudo and the palaeoclimatic and palaeoenvironmental conditions diachronically. In order to address this objective, we collected published and unpublished data from different sites on the eastern Iberian Peninsula. International audience; Estimating the genetic variation and demographic trends of species in decline is of major concern in conservation genetics. This study contributes to understanding how historical and anthropogenic factors shape the distribution of current genetic diversity in one of the most endangered reptiles in Western Europe, the Hermann's tortoise Testudo hermanni hermanni.
