Model and solution methods for some hub location problems

In this thesis we study some hub location problems in the context of transportation networks. These are combinatorial optimization problems appearing in situations where there is a need of transporting some traffic, like items, people, and information, from many origins to many destinations. Instead of sending these flows using a direct shipment between all pairs of nodes in the network, a subset of these nodes is selected to use as hubs, with the aim of consolidating and distribute the flows. Thus, hubs induce a subnetwork that sends the traffic more efficiently and at a cheaper cost, allowing economies of scale when large amounts of traffic between nodes on this subnet are transported. Adenosine-triphosphate-(ATP)-binding cassette (ABC) transport proteins are ubiquitously present membrane-bound efflux pumps that distribute endo- and xenobiotics across intra- and intercellular barriers. Discovered over 40 years ago, ABC transporters have been identified as key players in various human diseases, such as multidrug-resistant cancer and atherosclerosis, but also neurodegenerative diseases, such as Alzheimer���s disease (AD). Abstract Background A major problem of cancer treatment is the development of multidrug resistance (MDR) to chemotherapy. MDR is caused by different mechanisms such as the expression of the ABC-transporters P-glycoprotein (P-gp, MDR1, ABCB1) and breast cancer resistance protein (BCRP, ABCG2). Human ABCB1 confers cells resistance to cytotoxic guanidine alkaloids from Pterogyne nitens.

Multidrug resistance (MDR) caused by human ABCB1 (P-glycoprotein/MDR1) is one of the major obstacles in chemotherapy. To understand the mechanism of MDR by ABCB1 and circumvent the MDR, in the present study, we established human ABCB1-expressing cells (Flp-In-293/ABCB1 cells) and examined the cytotoxic effects of four guanidine alkaloids from Pterogyne nitens (galegine, nitensidine A, pterogynidine and pterogynine) using Flp-In-293/Mock and Flp-In-293/ABCB1 cells. The activity of ABCB1 in Flp-In-293/ABCB1 cells were confirmed by typical substrates for ABCB1 (taxol and vinblastine) in MTT assay.
