International genome-wide meta-analysis identifies new primary biliary cirrhosis risk loci and targetable pathogenic pathways

Primary biliary cirrhosis (PBC) is a classical autoimmune liver disease for which effective immunomodulatory therapy is lacking. Here we perform meta-analyses of discovery data sets from genome-wide association studies of European subjects (n=2,764 cases and 10,475 controls) followed by validation genotyping in an independent cohort (n=3,716 cases and 4,261 controls). We discover and validate six previously unknown risk loci for PBC (Pcombined<5 × 10−8) and used pathway analysis to identify JAK-STAT/IL12/IL27 signalling and cytokine–cytokine pathways, for which relevant therapies exist.

There are currently no biomarkers that predict hepatocellular carcinoma (HCC) risk in patients with hepatitis C virus (HCV)-related cirrhosis. We investigated the relationships among major histocompatibility complex (MHC) class I chain-related gene A (MICA) polymorphisms, plasma levels of soluble MICA (sMICA), and HCC risk in patients with HCV-related HCC. Coarse-Grained Barrier Trees of Fitness Landscapes

Recent literature suggests that local optima in fitness landscapes are clustered, which offers an explanation of why perturbation-based metaheuristics often fail to find the global optimum: they become trapped in a sub-optimal cluster. We introduce a method to extract and visualize the global organization of these clusters in form of a barrier tree. Barrier trees have been used to visualize the barriers between local optima basins in fitness landscapes. Our method computes a more coarsely grained tree to reveal the barriers between clusters of local optima. Abstract This study examines the acquisition of expertise in designing and developing information systems. The aim was to investigate how practical experience is related to contextual and strategic knowledge in problem-solving. Using a combination of expert–novice comparisons and longitudinal methods, professional systems analysts were compared with novices at the beginning and end of a seven month project-based course. The results show that during the course, the novices acquired a good deal of strategic competence in using domain-specific methods. Efficient image compression using directionlets

Directionlets are built as basis functions of critically sampled perfect-reconstruction transforms with directional vanishing moments imposed along different directions. We combine the directionlets with the space-frequency quantization (SFQ) image compression method, originally based on the standard two-dimensional wavelet transform. We show that our new compression method outperforms the standard SFQ as well as the state-of-the-art image compression methods, such as SPIHT and JPEG-2000, in terms of the quality of compressed images, especially in a low-rate compression regime. Objective To survey rheumatologists' preferences for the choice of a second-line disease-modifying antirheumatic drug (DMARD) after inadequate response with methotrexate (MTX) therapy in rheumatoid arthritis (RA). Methods Thirty-six rheumatologists stated their preferences for RA treatment after inadequate response with MTX therapy (optimal dose at least 6 months). From the initial scenario, we derived 54 vignettes varying by rheumatoid factor or anti–cyclic citrullinated peptide antibody presence, swollen joint count, Disease Activity Score in 28 joints, and structural damage. Power-aware design of MCML logarithmic adders

This paper describes the low-power design of a MOS current-mode logarithmic adder. The adder utilizes the Brent-Kung tree structure. The design strategy adopted is very simple and effective. Moreover, it can be utilized also for other types of logarithmic adders. To validate it, several adders were designed in a TSMC CMOS 130nm technology. Results of simulations indicate that the proposed methodology offers a good starting point before fine-tuning the design by SPICE simulations. Finally, the tradeoff that can be realized between performance and power consumption is discussed.
