Loop diuretics decrease the renal elimination rate and increase the plasma levels of trimethylamine‐N‐oxide

2018

Aims Trimethylamine-N-oxide (TMAO) is a novel cardiovascular risk marker. We explored the association of commonly used cardiovascular medications with TMAO levels in patients and validated the identified associations in mice. Methods Detailed history of drug treatment was recorded in 300 patients with cardiovascular disease without diabetes in an observational, cross-sectional study. Animal study was performed in CD1 mice. Results Median plasma TMAO (interquartile range) level was 2.144 (1.570-3.104) μmol l-1 . Among nine cardiovascular drug groups, the use of loop diuretics (0.510 ± 0.296 in users vs. 0.336 ± 0.272 in nonusers, P = 0.008) and mineralocorticoid receptor antagonists (0.482 ±…

Das System 2‐Halogenethylester/Cholinester als Zweistufen‐Schutzgruppe für die Carboxylfunktion von Aminosäuren und Peptiden

1979

Separation, isolation and stereochemical assignment of imazalil enantiomers and their quantitation in an in vitro toxicity test

2016

A simple method for the separation of the enantiomers of the fungicide imazalil was developed. Racemic imazalil was separated into its enantiomers with an enantiomeric purity of 99% using HPLC-UV with an enantioselective column (permethylated cyclodextrin) operated in reversed phase mode (water with 0.2% trimethylamine and 0.08% acetic acid and methanol). The absolute configuration of the separated enantiomers was assigned and unequivocally confirmed by optical rotation as well as by vibrational circular dichroism (VCD) and electronic circular dichroism (ECD) combined with ab-initio calculations. The same enantioselective column was also used to develop an HPLC-MS/MS method for the quantifi…

The Gut Microbial Metabolite Trimethylamine-N-Oxide Is Present in Human Cerebrospinal Fluid

2017

Trimethylamine-N-oxide (TMAO) is a small organic molecule, derived from the intestinal and hepatic metabolism of dietary choline and carnitine. Although the involvement of TMAO in the framework of many chronic diseases has been recently described, no evidence on its putative role in the central nervous system has been provided. The aim of this study was to evaluate whether TMAO is present at detectable levels in human cerebrospinal fluid (CSF). CSF was collected for diagnostic purposes from 58 subjects by lumbar puncture and TMAO was quantified by using liquid chromatography coupled with multiple-reaction monitoring mass spectrometry. The molecule was detected in all samples, at concentrati…

Abatement of 3-methylbutanal and trimethylamine with combined plasma and photocatalysis in a continuous planar reactor

2014

International audience; This paper deals with the 3-methylbutanal ((CH3)2CHCH2COH) removal with the help of a nonthermal surface plasma discharge coupled with photocatalysis. The capability of this process for gas treatment was studied. A planar reactor system was developed in order to perform the effect of adding photocatalytic material in plasma surface discharge barrier dielectric (SDBD) zone on (i) 3-methylbutanal removal, (ii) selectivity of CO2 and CO, (iii) byproducts formation such ozone formation. It was found that the influence of the UV light generated by SDBD reactor was very low. The activation of the photocatalyst media could be negligible. Whereas, the introduction of externa…

Diabetes is Associated with Higher Trimethylamine N-oxide Plasma Levels

2016

Recent studies have revealed strong associations between systemic trimethylamine N-oxide (TMAO) levels, atherosclerosis and cardiovascular risk. In addition, plasma L-carnitine levels in patients with high TMAO concentrations predicted an increased risk for cardiovascular disease and incident major adverse cardiac events. The aim of the present study was to investigate the relation between TMAO and L-carnitine plasma levels and diabetes. Blood plasma samples were collected from 12 and 20 weeks old db/db mice and patients undergoing percutaneous coronary intervention. Diabetic compared to non-diabetic db/L mice presented 10-fold higher TMAO, but lower L-carnitine plasma concentrations at 12 …

Characterization of a medium size Xe/TMA TPC instrumented with microbulk Micromegas, using low-energy gamma-rays

2014

NEXT-MM is a general-purpose high pressure (10 bar, $\sim25$ l active volume) Xenon-based TPC, read out in charge mode with an 8 cm $\times$8 cm-segmented 700 cm$^2$ plane (1152 ch) of the latest microbulk-Micromegas technology. It has been recently commissioned at University of Zaragoza as part of the R&D of the NEXT $0\nu\beta\beta$ experiment, although the experiment's first stage is currently being built based on a SiPM/PMT-readout concept relying on electroluminescence. Around 2 million events were collected during the last months, stemming from the low energy $\gamma$-rays emitted by a $^{241}$Am source when interacting with the Xenon gas ($\epsilon$ = 26, 30, 59.5 keV). The localized…

Oxygen Transfer from Trimethylamine N ‐Oxide to Cu I Complexes Supported by Pentanitrogen Ligands

2020

[N,N-bis(1-methyl-2-benzimidazolyl)methyl-N-(bis-2-pyridylmethyl)amine] (L1) and [N,N-bis(2-quinolylmethyl)-N-bis(2-pyridyl)methylamine] (L2) were employed to prepare CuII and CuI complexes for spectroscopic and structural characterization. [L1CuII(H2O)](NO3)2 and [L2CuII(NO3)]NO3 have Jahn–Teller distorted octahedral geometries and give rise to isotropic EPR spectra in frozen solution. [L1CuI(CH3CN)]OTf and [L2CuI(CH3CN)]OTf have distorted trigonal bipyramidal and tetrahedral solid-state structures, respectively. The N-donors display labile behavior in solution, based on variable-temperature 1H NMR studies. Addition of trimethylamine N-oxide (Me3NO) to solutions of [L1CuI(CH3CN)]OTf and [L…

Structure and Function of CutC Choline Lyase from Human Microbiota Bacterium Klebsiella pneumoniae.

2015

CutC choline trimethylamine-lyase is an anaerobic bacterial glycyl radical enzyme (GRE) that cleaves choline to produce trimethylamine (TMA) and acetaldehyde. In humans, TMA is produced exclusively by the intestinal microbiota, and its metabolite, trimethylamine oxide, has been associated with a higher risk of cardiovascular diseases. Therefore, information about the three-dimensional structures of TMA-producing enzymes is important for microbiota-targeted drug discovery. We have cloned, expressed, and purified the CutC GRE and the activating enzyme CutD from Klebsiella pneumoniae, a representative of the human microbiota. We have determined the first crystal structures of both the choline-…

Reductions of M{N(SiMe3)2}3 (M = V, Cr, Fe): Terminal and Bridging Low-Valent First-Row Transition Metal Hydrido Complexes and “Metallo-Transaminatio…

2021

The reaction of the vanadium(III) tris(silylamide) V{N(SiMe3)2}3 with LiAlH4 in diethyl ether gives the highly unstable mixed-metal polyhydride [V(μ2-H)6[Al{N(SiMe3)2}2]3][Li(OEt2)3] (1), which was structurally characterized. Alternatively, performing the same reaction in the presence of 12-crown-4 affords a rare example of a structurally verified vanadium terminal hydride complex, [VH{N(SiMe3)2}3][Li(12-crown-4)2] (2). The corresponding deuteride 2D was also prepared using LiAlD4. In contrast, no hydride complexes were isolated by reaction of M{N(SiMe3)2}3 (M = Cr, Fe) with LiAlH4 and 12-crown-4. Instead, these reactions afforded the anionic metal(II) complexes [M{N(SiMe3)2}3][Li(12-crown-…
