Real-time Fluorescence Measurement of Enterovirus Uncoating

2019

Viruses need to open, i.e., uncoat, in order to release their genomes for efficient replication and translation. Especially for non-enveloped viruses, such as enteroviruses, the cues leading to uncoating are less well known. The status of the virus has previously been observed mainly by transmission electron microscopy using negative staining, cryo electron microscopy, X-ray crystallography or gradient separation (reviewed in Tuthill et al., 2010, Myllynen et al., 2016, Ruokolainen et al., 2019). However, monitoring of uncoating has been limited by the lack of methods detecting dynamic changes of the virions. Here, we present a real-time fluorescence based protocol, which detects the viral …

Axial dispersion model for solid flow in liquid suspension in system of two mixers in total recycle

2006

The measurement of residence time distribution of solid particles in solid-liquid suspension is experimentally difficult. However, the twin system approach is particularly suited for the assessment of particle RTD in flow systems as it allows overcoming some of the usual difficulties generally encountered in this kind of measurement. Twin system consists of two vessels and external piping in total recycle. Experimental results from this system can be evaluated using Z-transforms to derive particle RTD for subsequent testing of alternative flow models. Recently, the axial dispersion model was applied using the "advection diffusion equation" (sometimes called the"diffusion with bulk flow equa…

Automatic analysis of speckle photography fringes

1997

Speckle interferometry is a technique adequate to metrological problems such as the measurement of object deformation. An automatic system of analysis of such measurements is given; it consists of a motorized x-y plate positioner controlled by computer, a CCD video camera, and software for image analysis. A fringe-recognition algorithm determines the spacing and orientation of the fringes and permits the calculation of the magnitude and direction of the displacement of the analyzed object point in images with variable degrees of illumination. For a 256 x 256 pixel image resolution, the procedure allows one to analyze from three fringes to a number of fringes that corresponds to 3 pixels/fri…

Maximum likelihood for target location in the presence of substitutive noise .

2001

We consider the optimal likelihood algorithm for the estimation of a target location when the images are corrupted by substitutive noise. We show the relationship between the optimal algorithm and the sliced orthogonal nonlinear generalized (SONG) correlation. The SONG correlation is based on the application of a linear correlation to corresponding binary slices of both the input scene and the reference object with appropriate weight factors. For a particular case, we show that the optimal strategy is a function of only the number of pixels for which the gray values in the noisy image match the ones of the reference image when the substitutive noise is uniformly distributed. This is exactly…

Cognitive structuring and placebo effect

2017

Abstract In much of the research concerning the placebo phenomenon, the idea that placebo effects may vary in strength depending on a patient's personal characteristics or traits has been investigated. Findings regarding possible personality differences in placebo response, however, are conflicting and non-systematic. In this article a new theoretical attempt to explain the placebo phenomenon is offered. The authors postulate that the power of the placebo effect is moderated by the extent of use of cognitive structuring, which in turn is influenced by the interaction between the individuals' level of need for cognitive closure and their ability to achieve this state. To test this assumption…

On the hardness of evaluating criticality of activities in a planar network with duration intervals

2003

Complexity results for problems of evaluating the criticality of activities in planar networks with duration time intervals are presented. We show that the problems of asserting whether an activity is possibly critical, and of computing bounds on the float of an activity in these networks are NP-complete and NP-hard, respectively.

Assessment of tools for urban energy planning

2019

Abstract In recent years, studies and policies have encouraged the diffusion of distributed energy supply technologies and large integration of renewable sources. This increases the need for new professionals in energy planning. Several computational tools for energy planning have been developed as described in the technical literature. However, energy planners of urban/district areas engaged in the transition towards smart systems related to buildings energy services require well-documented tools to evaluate the combination of available energy sources by proper conversion technologies. With this, after a scientific review, we selected 17 tools targeted on an urban/districts scale that can …

Identification and quantification of meat product ingredients by whole-genome metagenomics (All-Food-Seq)

2019

AbstractComplex food matrices bear the risk of intentional or accidental admixture of non-declared species. Moreover, declared components can be present in false proportions, since expensive taxa might be exchanged for cheaper ones. We have previously reported that PCR-free metagenomic sequencing of total DNA extracted from sausage samples combined with bioinformatic analysis (termed All-Food-Seq, AFS), can be a valuable screening tool to identify the taxon composition of food ingredients. Here we illustrate this principle by analysing regional Doner kebap samples, which revealed unexpected and unlabelled poultry and plant components in three of five cases. In addition, we systematically ap…
