Neurogenic Inflammation in Complex Regional Pain Syndrome (CRPS)

This chapter explains symptoms and nature of neurogenic inflammation and its importance in posttraumatic complex regional pain syndrome (CRPS). Neurogenic inflammation regularly accompanies excitation of primary afferent nociceptors. It has two major components – plasma extravasation and vasodilatation. The most important mediators are the neuropeptides calcitonin gene-related peptide (CGRP) and substance P (SP). After peripheral trauma, immune reaction (e.g., cytokines) and the attempts of the tissue to regenerate (e.g., growth factors) sensitize nociceptors and thereby amplify neurogenic inflammation. Tumour-cell-endothelial interactions: free radicals are mediators of melanoma-induced endothelial cell damage

Damage to vascular endothelium may play an important role during metastasis. We used a three-dimensional model of tumour cell extravasation to test the hypothesis that certain types of tumour cells are able to induce vascular endothelial cell injury. Multicellular tumour spheroids (MCTS) of 14 human cancer cell lines and spheroids from two benign cell lines were transferred onto confluent monolayers of human endothelial cells (EC). MCTS from 4 of 7 melanoma cell lines induced damage of the endothelium which was closely associated with tumour cell attachment. The mode of lymphocyte transendothelial migration in the postcapillary high endothelial venules (HEVs) of Peyer's patches during normal homing and acute inflammation in the guinea pig was studied. Sodium/hydrogen exchange inhibition with cariporide reduces leukocyte adhesion via P-selectin suppression during inflammation

Background and purpose: The Na+/H+ exchange (NHE) inhibitor cariporide is known to ameliorate ischaemia/reperfusion (I/R) injury by reduction of cytosolic Ca2+ overload. Leukocyte activation and infiltration also mediates I/R injury but whether cariporide reduces I/R injury by affecting leukocyte activation is unknown. Glutathione in Cancer Biology and Therapy

The glutathione (GSH) content of cancer cells is particularly relevant in regulating mutagenic mechanisms, DNA synthesis, growth, and multidrug and radiation resistance. In malignant tumors, as compared with normal tissues, that resistance associates in most cases with higher GSH levels within these cancer cells. Thus, approaches to cancer treatment based on modulation of GSH should control possible growth-associated changes in GSH content and synthesis in these cells.
