DNA methylation episignature testing improves molecular diagnosis of Mendelian chromatinopathies

2021

Abstract Purpose Chromatinopathies include more than 50 disorders caused by disease-causing variants of various components of chromatin structure and function. Many of these disorders exhibit unique genome-wide DNA methylation profiles, known as episignatures. In this study, the methylation profile of a large cohort of individuals with chromatinopathies was analyzed for episignature detection. Methods DNA methylation data was generated on extracted blood samples from 129 affected individuals with the Illumina Infinium EPIC arrays and analyzed using an established bioinformatic pipeline. Results The DNA methylation profiles matched and confirmed the sequence findings in both the discovery an…

Transition from self tilt to object tilt during maintained lateral tilt in parabolic flight.

1991

Abstract 19 young healthy subjects were subjected to parabolic rollercoaster flight. A horizontal luminous line was seen by the subjects in a headfixed goggle device. During the hypergravic phases of parabolic flight the luminous line seemed to rotate into and during the hypogravic phase against the direction of static head tilt. Ocular counter rotation and activity of the neck position receptors cannot explain these subjective rotations. We conclude that information from the otolith system, converging with visual information within the brain, dislocated the headfixed visual target line. While the retinal image of the luminous line remains unchanged, loading and unloading the otoliths in pa…

Is backward disequilibrium in the elderly caused by an abnormal perception of verticality? A pilot study

2007

International audience; OBJECTIVE: We hypothesised that backward disequilibrium (BD), defined by a posterior position of the centre of mass with respect to the base of support, could be caused by a backward tilt in the perception of verticality. METHODS: The relationship between BD, the perception of verticality, and the history of falls in 25 subjects aged 84.5+/-7.4 years was analysed. An original ordinal scale, the BD scale (BDS), was used to quantify BD. Postural (PV) and haptic verticals (HV) were measured in sagittal plane. RESULTS: BDS scores closely correlated with the number of falls (r = 0.81, p =10(-5)). The more the PV was tilted backward, the greater the BDS scores (r = -0.95, …

Vertigo in childhood: A methodological approach

2012

Children complaints dizziness merit meticulous evaluation to differentially diagnose a vestibular disease. A syndrome mimicking certain classic signs and symptoms of adult vestibular disorders may be presents in children, such as benign paroxysmal positional vertigo, usually associated with aging. Benign paroxysmal vertigo, in which migraine is a manifestation, with sudden onset of dizziness is a rare peripheral vestibular disorder that is commonly ignored or misdiagnosed. This review covers the development of the diagnosis, evaluation and treatment approaches of vertigo of childhood, a valid support for physician that approach dizzy children (Ref. 25).

Firing characteristics of vestibular nuclei neurons in the alert monkey after bilateral vestibular neurectomy

1992

After destruction of the peripheral vestibular system which is not activated by moving large-field visual stimulation, not only labyrinthine-ocular reflexes but also optokinetic-ocular responses related to the "velocity storage" mechanism are abolished. In the normal monkey optokinetic-ocular responses are reflected in sustained activity changes of central vestibular neurons within the vestibular nuclei. To account for the loss of optokinetic responses after labyrinthectomy, inactivation of central vestibular neurons consequent on the loss of primary vestibular activity is assumed to be of major importance. To test this hypothesis we recorded the neural activity within the vestibular nuclea…

Estudio genético molecular del síndrome de Usher en España

2005

Usher syndrome (USH) associates deafness and retinitis pigmentosa (RP). It is a disease both clinically and genetically heterogeneous. It is inherited as an autosomal recessive trait and its prevalence makes it the most frequent association of hearing loss and RP. Clinically Usher syndrome is divided into type I (USH1), II (USH2) and III (USH3), according to the severity of hearing loss, age of onset of RP and the existence or not of vestibular dysfunction. There are at least 7 different localizations for USH1 and 5 genes have been identified. For USH2, 3 loci and 2 genes have been reported and USH3 is due to Clarin-1 gene. Our aim is to perform a clinical and genetic characterization of al…

European vestibular experiments on the Spacelab-1 mission: 2. Experimental equipment and methods

1986

A series of vestibular experiments were performed in conjunction with the first Spacelab mission, consisting of sets of pre-, in- and postflight tests. A multipurpose experimental apparatus used for the diverse flight and ground tests is presented. Additional apparatus together with the multi-purpose package were used in the baseline data collection facility at the landing site at NASA Dryden Flight Research Facility for the ground tests. The tests involved optokinetic, caloric and mechanical (whole-body or head-alone) stimulation. The latter included linear acceleration in the subject's x, y and z axes, static roll and yaw about an earth-vertical axis. Physiological parameters such as elec…

Influence of interaction techniques o n VIMS in virtual environments : estimation and prediction

2014

Understanding oculo-vestibular dynamics during sensory rearrangement in the central nervous system plays an extremely important role in better understanding human perception, and improves the technology in many engineering fields. Besides, the sensory conflict that occurs between ocular, vestibular and proprioception during sensory rearrangement at certain occasions might adversely affect the user performance in a wide variety of domains including flight/car simulators, scale-one 3D systems, large-scale displays, serious games, and so on. Therefore, knowing the condition in which the sensory conflict happens has a great deal of importance. This study aims at understanding the nature of sens…
