Microglia and α-synuclein implication in Parkinson's disease
2011
Age-related neurodegenerative disorders like Parkinson’s disease take an enormous toll on individuals and on society. Despite extensive efforts, Parkinson’s disease remains incurable and only very limited treatments exist. Indeed, Parkinson’s pathogenesis is still not clear and research on its molecular mechanisms is ongoing. In this study, we focused our interest on two abnormal events occurring in Parkinson’s patients, namely α-synuclein aggregation and microglial activation. We first investigated α-synuclein and its abnormal polymerisation. For this purpose, we developed novel methods, which allowed the in vitro production of different types of α-synuclein oligomers. Using highly sensiti…
In vitro biotechnology approaches now available for ‘beautiful’ vavilovia (Vavilovia formosa)
2014
BAP Pôle GEAPSI; Efficient in vitro propagation of Vavilovia formosa, plant regeneration from callus, protoplast isolation and culture to differentiated callus of V.formosa were developed and its relative nuclear DNA content by flow cytometry was established. The summation of biotechnology tools now available should foster evolutionary studies on the tribe Fabeae and, intime, V.formosa could be come a potential source of novel agronomic adaptive traits.

Genetic plasticity of atrazine-degrader: evidence for the deletion of atzABC genes in pseudomonas sp. adp in response to cyanuric acid selection pres…
2010
International audience; Several bacterial strains are known to metabolize s-triazines herbicides among which atrazine reported worldwidel to contaminate soil and water resources. The complete mineralization of atrazine to carbon dioxide and ammonium is catalyzed by 6 hydrolytic enzymes encoded by atzABCDEF genes located on the plasmid pADP1 in Pseudomonas ADP. The atzABC genes code for the upper pathway transforming atrazine to cyanuric acid, while the atzDEF genes code for the lower pathway transforming cyanuric acid to simple compounds. In order to study the genetic plasticity of the atrazine catabolic pathway, we performed an in vitro evolution study consisting in applying a continuous s…

Robust Automated Assessment of Human Blastocyst Quality using Deep Learning
2018
AbstractMorphology assessment has become the standard method for evaluation of embryo quality and selecting human blastocysts for transfer inin vitro fertilization(IVF). This process is highly subjective for some embryos and thus prone to human bias. As a result, morphological assessment results may vary extensively between embryologists and in some cases may fail to accurately predict embryo implantation and live birth potential. Here we postulated that an artificial intelligence (AI) approach trained on thousands of embryos can reliably predict embryo quality without human intervention. To test this hypothesis, we implemented an AI approach based on deep neural networks (DNNs). Our approac…
Larval zebrafish as an in vitro model for evaluating toxicological effects of mycotoxins.
2020
The presence of mycotoxins in food has created concern. Mycotoxin prevalence in our environment has changed in the last few years maybe due to climatic and other environmental changes. Evidence has emerged from in vitro and in vivo models: some mycotoxins have been found to be potentially carcinogenic, embryogenically harmful, teratogenic, and to generate nephrotoxicity. The risk assessment of exposures to mycotoxins at early life stages became mandatory. In this regard, the effects of toxic compounds on zebrafish have been widely studied, and more recently, mycotoxins have been tested with respect to their effects on developmental and teratogenic effects in this model system, which offers …
