Palbociclib is an oral CDK4/6 inhibitor indicated in HR+/HER2- advanced or metastatic breast cancer in combination with hormonotherapy. Its main toxicity is neutropenia. The aim of our study was to describe the kinetics of circulating neutrophils from real-life palbociclib-treated patients. A population pharmacokinetic (popPK) model was first constructed to describe palbociclib pharmacokinetic (PK). Individual PK parameters obtained were then used in the pharmacokinetic/pharmacodynamic (PK/PD) model to depict the relation between palbociclib concentrations and absolute neutrophil counts (ANC). What really causes large price changes?

We study the cause of large fluctuations in prices in the London Stock Exchange. This is done at the microscopic level of individual events, where an event is the placement or cancellation of an order to buy or sell. We show that price fluctuations caused by individual market orders are essentially independent of the volume of orders. Instead, large price fluctuations are driven by liquidity fluctuations, variations in the market's ability to absorb new orders. Even for the most liquid stocks there can be substantial gaps in the order book, corresponding to a block of adjacent price levels containing no quotes. In this thesis, we present an numerical study of turbulent forced convection in a heat sink provided with a transverse baffle in the bypass. The first model is composed of plates fins and the second consists of adding pin fins between the plates fins. The governing equations, based on the k- SSt turbulence model, are disscredized and solved by the finite volume method and the SIMPLE algorithm. This paper presents a simulation tool to determine the structural deflections and corresponding leakage flow in a hydrostatic displacement motor. The simulation tool is applied to a new motor principle that is categorized as an extreme low-speed high-torque motor with dimensions that calls for attention to the volumetric efficiency. To counteract structural deflections the motor is equipped with compensation pressure volumes that may be used to limit the leakage flow across the end faces of the circular rotor. ABSTRACT The combined effects of subzero temperature and high pressure on the inactivation of Escherichia coli K12TG1 were investigated. Cells of this bacterial strain were exposed to high pressure (50 to 450 MPa, 10-min holding time) at two temperatures (−20°C without freezing and 25°C) and three water activity levels (a w ) (0.850, 0.992, and ca. 1.000) achieved with the addition of glycerol. There was a synergistic interaction between subzero temperature and high pressure in their effects on microbial inactivation.
