Improved Mesophyll Protoplast Culture and Plant Regeneration in Arabidopsis thaliana (L.) Heynh., Genotype Landsberg Erecta

Summary The response of in vitro cultured Arabidopsis thaliana mesophyll protoplasts was investigated with the aim of characterising the crucial factors that affect plating efficiencies and plant regeneration. We selected the genotype Landsberg erecta, which is frequently used for genetic studies, but which is known to respond poorly in protoplast culture. Factors permitting vigorous vegetative growth of the donor plants, i.e. short light period and avoidance of high temperature, were found to be advantageous. Abstract In the last decade, high-density olive planting systems have been developed to facilitate less costly mechanical harvesting and pruning by using straddle harvesters. High-density orchards allow faster production since the first crop of fruit starts a few years after planting. In high-density orchards the control of tree size is essential. Activities of NAD(+)-dependent sorbitol dehydrogenase (SDH), sorbitol oxidase (SOX), sucrose synthase (SS), acid invertase (AI), and neutral invertase (NI) in 'Encore' peach (Prunus persica L.) fruits and developing shoot tips were assayed during the growing season to determine whether carbohydrate metabolizing enzymes could serve as indicators of sink strength. Early bearing and vegetative growth of 153 Sicilian olive accessions

In 2014, an olive germplasm collection was established in south-western Sicily, Italy. The collection included 153 Sicilian accessions and 3 international varieties (reference), each with 7 trees, spaced at 5×2 m and trained to hedgerow system. The aim of the trial was to study early growth and yield responses of the accessions to super-high planting density (SHD). Trunk cross sectional area, tree height, pruning weights, shoot positioning, blooming and fruit yield (kg tree-1) were measured in 2015 and 2016. After a brief summary of the systematics of the Vitaceae the question of the nomenclature of cultivated grapevines is discussed. Intra-varietal variability is discussed using morphological and molecular characteristics. EFFECT OF DIFFERENT ROOTSTOCKS ON VEGETATIVE GROWTH OF JAPANESE AND EUROPEAN PLUM CULTIVARS IN SOUTHERN ITALY: PRELIMINARY RESULTS

The preliminary results of a new rootstock trial for Japanese and European plum cultivars are reported. Two clonal Prunus cerasifera ('Myrobalan 29C' and 'MrS 2/5'), two Prunus domestica ('Penta' and 'Tetra') and the peach × almond 'GF 677' were tested in a warm and dry climate of Southern Italy where the plum industry is recently increasing, particularly for the Japanese type. 'GF 677' showed the most vigorous habit together with a higher production at the first bearing year in comparison with 'Myrobalan 29C', while the two P. domestica types resulted in very low vigour. Specific rootstocks can differentially influence the vegetative growth and development of fruit trees. However, the physiological mechanism involved in this phenomenon has been elusive. Cutting Type and IBA Treatment Duration Affect <i>Teucrium fruticans</i> Adventitious Root Quality

Root development of stem cuttings of Silver Germander (Teucrium fruticans) was investigated in relation to cutting type and indole-3-butyric acid (IBA) treatment. Terminal cuttings of a clone grown in Sicily were trimmed to three types: terminal cuttings with apex (TWA), terminal cuttings without apex (TWOA) or sub-terminal cuttings (ST). To verify the cutting response to exogenous auxin, cuttings were dipped to a 2.0 cm depth in a 0.5% indole-3-butyric acid solution for 0, 5 or 7 minutes. Overall percent survival was 97 to 98%.
