The sea urchin sns5 chromatin insulator settles a gene therapy vector into an independent domain of expression in the vertebrate genome

One of the critical aspects of introducing a transgene into the eukaryotic genome is the great variability of gene expression due to position effects (1). Chromatin-dependent repressive states could be overcome by incorporation in the transgene of chromatin insulators, functioning to establish and delimit domains of expression. We have previously demonstrated that the sea urchin sns5 DNA element has the typical features of an insulator: by acting as enhancer blocker, it shields promoters from neighboring regulatory elements, and by acting as barrier it buffers a transgene from the propagation of condensed chromatin (2,3). Gli autori hanno studiato le modificazioni morfologiche delle apofisi trasverse della settima vertebra cervicale. Sono state osservate principalmente due variazioni: la mega - apofisi trasversa e la costa cervicale. Gli autori hanno riscontrato una incidenza di variazioni notevolmente superiore rispetto a quanto riportato in letteratura, suppongono che ciò sia dovuto alla relativa > geografica e socio - economica ed ipotizzano una trasmissione genetica di tale variazione del tipo autosomico recessivo.

Treatment adherence and QoL with intermittent PTH therapy: Results from a multicentre Italian study after 6 months

This ongoing multicentre study was carried out to investigate the adherence to intermittent parathyroid hormone treatment and the effect on QoL in a typical Italian population eligible for drug reimbursement (i.e. a new vertebral or hip fracture following at least one year of anti-resorptive therapy or three severe vertebral fractures or two severe and one femur fracture). 204 female patients (mean age: 72.6±8.3) were enrolled between March 2008 and April 2009. These patients were treated with PTH (PTH 1–34 or PTH 1–84) according to Italian legislation. Here, we report the results obtained at month 6.

L'importanza dell'esercizio terapeutico nel trattamento integrato dell'osteoporosi vertebrale deriva dal fatto che produce sul tessuto osseo effetti metabolici positivi. Scopo del rpogramma chinesiterapico è di ottenere un buon tonotrofismo dei musocli deficitari e rilassare quelli che per compenso sono in uno stato di contrattura permanente e quindi dolorosa. Il compito del fisiatra deve indirizzarsi verso l'istruzione e l'adattamento della vita personale e di relazione del soggetto osteoporotico.
