This paper describes a work aimed at improving the conditions of an extraction method, coupling GC-MS determination, for the analysis without cleanup phase, of polycyclic aromatic hydrocarbons (PAHs) from sediment samples. The automatic Soxhlet extraction in warm mode (using Extraction System B-811 Standard, Buchi) has demonstrated advantages for automation, reduced extraction time, and lower solvent use than for conventional Soxhlet extraction. Under these conditions, the recoveries are very good as they resulted greater than 85 % and, in most of the cases, near 100 %. A fast, sensitive and cheap procedure has been developed for the determination of inorganic mercury (i-Hg) and organic mercury (o-Hg) in mushroom samples. The procedure is based on the use of cold vapour atomic fluorescence spectrometry (CV-AFS). The method involves the extraction of total mercury (t-Hg) with diluted HCl, followed by measurements of the corresponding Hg vapour under two different conditions: (i) directly to determine i-Hg, and (ii) after oxidation with a mixture KBr/KBrO3 to determine t-Hg. o-Hg was estimated from the difference between t-Hg and i-Hg. Abstract Adsorbents for high-performance affinity chromatography were prepared by bonding proteins and reactive Procion triazine dyes to 3-isothiocyanatopropyl- and 3-aminopropylsilicas. The materials prepared were used successfully in the separation of hydrophobic plasma membrane proteins of cultured human fibroblasts.
