Protease-mediated processing of Argonaute proteins controls small RNA association

SummarySmall RNA pathways defend the germlines of animals against selfish genetic elements and help to maintain genomic integrity. At the same time, their activity needs to be well-controlled to prevent silencing of ‘self’ genes. Here, we reveal a proteolytic mechanism that controls endogenous small interfering (22G) RNA activity in the Caenorhabditis elegans germline to protect genome integrity and maintain fertility. We find that WAGO-1 and WAGO-3 Argonaute (Ago) proteins are matured through proteolytic processing of their unusually proline-rich N-termini. A partir del estudio de la obra teatral Salomé (1893) de Oscar Wilde, comprobamos cómo los diferentes directores del cine mudo de finales del siglo XIX y principios del siglo XX (hasta los años 20) se han influido de esta obra, y analizamos si ha sido una copia literal o en cambio si ha sido una influencia mucho más sutil. Cellular UDP-Glucose Deficiency Caused by a Single Point Mutation in the UDP-Glucose Pyrophosphorylase Gene

We previously isolated a mutant cell that is the only mammalian cell reported to have a persistently low level of UDP-glucose. In this work we obtained a spontaneous revertant whose UDP-glucose level lies between those found in the wild type and the mutant cell. The activity of UDP-glucose pyrophosphorylase (UDPG:PP), the enzyme that catalyzes the formation of UDP-glucose, was in the mutant 4% and in the revertant 56% of the activity found in the wild type cell. Sequence analysis of UDPG: PP cDNAs from the mutant cell showed one missense mutation, which changes amino acid residue 115 from glycine to aspartic acid. Abstract We have previously isolated a fibroblast mutant cell with high resistance to the two Rho-modifying glucosyltransferase toxins A and B of Clostridium difficile. We demonstrate here a low level of UDP-glucose in the mutant, which explains its toxin resistance since: (i) to obtain a detectable toxin B-mediated Rho modification in lysates of mutant cells, addition of UDP-glucose was required, and it promoted the Rho modification dose-dependently; (ii) high pressure liquid chromatography analysis of nucleotide extracts of cells indicated that the level of UDP-glucose in the mutant (0.8 nmol/106 cells) was lower than in the wild type (3.7 nmol/106 cells); and (iii) sensitivity to toxin B…

This introduction explicates the central issues informing the chapters in the volume. We outline the epistemological development of Second Language Acquisition research as it has evolved from being predominantly individual-cognitive to a more pluralistic endeavor in which social approaches to cognition and learning are becoming central. Social interaction has been recognized as key to language learning since the 1970’s but the field is still lacking in research that studies the everyday social-interactional ecology in which the L2 speaker acts. The environmental and economic costs of greenery depend on the planning criteria adopted and on the plants used. These costs can become more sustainable and can also be significantly lowered by using native flora and getting inspired by local plant communities. Nature-based solutions entail the use of species that co-occur naturally, thus replicating a model of coexistence consolidated by the evolutionary coherence of the biosphere. CITES konvencijas noteikumu pārkāpumu izpēte starptautiskajā lidostā “Rīga”

Maģistra darba mērķis ir izanalizēt CITES konvencijas pārkāpumus starptautiskajā lidostā „Rīga” un visā Latvijas teritorijā, to cēloņus, izstrādāt ieteikumus un risinājumus šo pārkāpumu novēršanai. Darbā analizēti autores veikto aptauju rezultāti, intervējot ielidojošos pasažierus starptautiskajā lidostā „Rīga” no Eiropas Savienības un trešajām valstīm par attieksmi pret CITES konvenciju, noskaidrojot informētības līmeni un informācijas avotus. Izanalizēta CITES konvencijas pārkāpumu skaita dinamika lidostā „Rīga” periodā no 2011.-2015.gadam. Maģistra darbs sastāv no 66 lapaspusēm. Darbs satur 23 attēlus, 3 tabulas, 54 izmantotos literatūras avotus un 1 pielikumu.

Background & Aims: A novel family of proton-sensing G-proteinâcoupled receptors, including ovarian cancer G-proteinâcoupled receptor 1 (OGR1) (GPR68) has been identified to play a role in pH homeostasis. Hypoxia is known to change tissue pH as a result of anaerobic glucose metabolism through the stabilization of hypoxia-inducible factor-1α. We investigated how hypoxia regulates the expression of OGR1 in the intestinal mucosa and associated cells.
