Synthesis of novel polypyridylcarbonylpyridines from triazolopyridines. Building blocks in supramolecular chemistry

The synthesis from triazolopyridines 1, of novel triazolopyridylcarbonylpyridylcarbonyltriazolopyridines (TPyCOPyCOTPy) (tpcpctp) 7, and polypyridylcarbonylpyridines (pPyCOPy) (ppcp) 14, building blocks in supramolecular chemistry, is described. These compounds are interesting polynitrogenated ligands as potential molecular sensors, new magnetic materials, single molecular magnets, or in the emerging science of nanomaterials. Preclinical and clinical evidence of activity of pazopanib in solitary fibrous tumour

Abstract Background To explore the activity of pazopanib in solitary fibrous tumour (SFT). Patients and methods In a preclinical study, we compared the activity of pazopanib, sorafenib, sunitinib, regorafenib, axitinib and bevacizumab in a dedifferentiated-SFT (DSFT) xenotransplanted into Severe Combined Immunodeficiency (SCID) mice. Antiangiogenics were administered at their reported optimal doses when mean tumour volume (TV) was 80 mm3. Drug activity was assessed as TV inhibition percentage (TVI%). Progettazione e sintesi di nuovi derivati 4-chinazolinonici potenziali inibitori della diidrofolato reduttasi

I chinazolinoni sono composti eterociclici azotati che, insieme alle chinazoline, rappresentano degli importanti farmacofori in possesso di un ampio spettro di proprietà biologiche tra cui quella antitumorale. Recentemente sono stati riportati in letteratura dei derivati 4-chinazolinonici in grado di inibire in vitro l’enzima diidrofolato reduttasi (DHFR) con IC50 comprese tra 0.4 e 1.0 µM [1]. Allo scopo di progettare la sintesi di nuovi potenziali inibitori della DHFR, è stato condotto uno studio di modellistica molecolare considerando tale enzima come biotarget. A series of 1:1 adducts of the type [(L)R(n)SnX(4-n)] . zH(2)O (L = bis(4-methylpyrazol-1-yl)methane (L(4)), bis(3,4,5-trimethylpyrazol-1-yl)methane (L(T)), 1,2-bis(pyrazol-1-yl)ethane (L(A)) or 1,2-bis(3,5-dimethylpyrazol-1-yl)ethane (L(B)); R = Me, Et, Bu or Ph; X = I, Br or Cl; n = 0, 1 or 2; z = 1, 1.5 or 2), and the likely polynuclear [(L(A))(5)(SnCl4)(4)] . Abstract Ionic liquids are dual modifiers composed by a large anion and a large cation, which interact with both the hydrophobic alkyl-bonded phase and the anionic residual silanols in C18 columns. In reversed-phase liquid chromatography in the absence of additives, cationic basic compounds give rise to broad and asymmetrical peaks as a result of ionic interactions with residual free silanols on silica-based stationary phases. Ionic liquids (ILs), added to the mobile phase, have been suggested as alternatives to amines to block the activity of silanols.
