Estudio del mecanismo de reacción e inhibición de cisteína proteasas mediante métodos multiescala

En esta tesis doctoral se realizaron estudios computacionales para las enzimas caspasa-1 y la 3CLpro del SARS-CoV-2. Estas enzimas pertenecen a la familia de las cisteína proteasas, las cuales hidrolizan sus respectivos sustratos rompiendo un enlace peptídico en particular. En esta reacción interviene una diada catalítica conformada por un par Cis-His. La caspasa-1 fue seleccionada debido a su importancia farmacológica en la enfermedad de Alzheimer, y la enzima 3CLpro del SARS-CoV-2 por estar involucrada en el proceso de replicación del virus responsable de la enfermedad del COVID-19. Summary: Chromatin organization plays a crucial role in tissue homeostasis. Heterochromatin relaxation and consequent unscheduled mobilization of transposable elements (TEs) are emerging as key contributors of aging and aging-related pathologies, including Alzheimer’s disease (AD) and cancer. However, the mechanisms governing heterochromatin maintenance or its relaxation in pathological conditions remain poorly understood. Here we show that PIN1, the only phosphorylation-specific cis/trans prolyl isomerase, whose loss is associated with premature aging and AD, is essential to preserve heterochromatin. Evaluation of the animal-assisted therapy in Alzheimer's disease

Animal-assisted therapy sessions have been set up in a protected unit for patients with a dementia-related syndrome. The aim is to measure the effects of animal-assisted therapy on behavioural disorders in daily life and care. The results obtained provided some interesting areas to explore and recommendations with a view to optimising the implementation of such a system.

Lexical Substitution and Paraphasia in Advanced Dementia of the Alzheimer Type

Abstract The paper presents the case study of lexical selection in Alzheimer-type dementia. Lexical substitutions in poem recitation and conversations of a Russian speaker, who suffered Alzheimer-type dementia, were analyzed on the background of the lexical retrieval and slip-of-the-tongue phenomena. The classification of the substitutions is worked out on the basis of the links between a target word and its substitutions. The current context plays an essential role as natural priming for a substitution in a poem recitation. Tutkimukseni kuvaa työikäisenä etenevään muistisairauteen sairastuneiden ihmisten elä-mää. Tutkimukseni tavoitteena on tuottaa tietoa työikäisenä muistisairauteen sairastunei-den ihmisten arjesta sekä määrittää sairastuneiden sosiaalisen kuntoutuksen elementtejä. Tutkimuksen päämääränä on nostaa esille sairastuneiden oma ääni ja kokemuksellinen asiantuntijuus. Tutkimuksen metodologisena lähestymistapana on narratiivisuus. Aineiston keruumene-telmänä käytetään narratiivista haastattelua. Haastatteluihin osallistuu viisi Alzheimerin tautiin sairastunutta ihmistä.
