A rapid method is described for the identification and determination of amphetamine and methamphetamine in human urine samples by liquid chromatography with UV-Vis detection. The samples were transferred onto a C18 solid-phase extraction column and chromatographed on a Hypersil ODS RP C18, 5 microns (250 x 4 mm I.D.) with an acetonitrile-water elution gradient containing propylamine. Under these conditions, the amines are eluted with a short retention time. The procedure has been applied to the determination of amphetamine and methamphetamine in the range 0.3-4.0 micrograms/ml in spiked urine samples. Automated Determination of Trimipramine and N-Desmethyl-Trimipramine in Human Plasma or Serum by HPLC With On-Line Solid Phase Extraction

A fully automated method including column-switching and isocratic high performance liquid chromatography (HPLC) was developed for determination of the tricyclic antidepressant trimipramine (T) and its N-demethylated metabolite N-desmethyltrimipramine (DT). The limit of quantification was below 10 ng/ml for T and DT. The assay revealed linearity between detector response and drug concentration in a therapeutically relevant range of 10 to 500 ng/ml. The mean intra- and interassay variabilities were 6.2 and 12.3%, respectively, for T and 4.7 and 8.7% respectively, for DT. Derivatization of tertiary amphetamines with 9-fluorenylmethyl chloroformate for liquid chromatography: determination of N-methylephedrine.

The fluorogenic reagent 9-fluorenylmethyl chloroformate (FMOC) was evaluated for the derivatization of tertiary amphetamines prior to liquid chromatographic analysis. Conditions for the derivatization were investigated, including the reaction time, the derivatization reagent concentration and the pH, using N-methylephedrine as a model compound. On the basis of these studies, a method for the quantification of N-methylephedrine is presented. Quantitation of talinolol in rat plasma by LC-MS-MS.

The aim of this study was to develop and validate an assay based on liquid chromatography-tandem mass spectrometry to quantitate talinolol in rat plasma. After a simple protein precipitation step, separation was performed by reversed-phase liquid chromatography using gradient elution with acetonitrile-water-formic acid. Electrospray ionization in the positive ion mode with multiple reaction monitoring was used to analyze talinolol employing propranolol as internal standard. The calibration curve for talinolol was linear over the concentration range 1-250 ng/mL with a correlation coefficient0.995. Determination of mercury in rice by cold vapor atomic fluorescence spectrometry after microwave-assisted digestion.

Abstract A cold vapor atomic fluorescence spectrometry method (CV-AFS) has been developed for the determination of Hg in rice samples at a few ng g−1 concentration level. The method is based on the previous digestion of samples in a microwave oven with HNO3 and H2O2 followed by dilution with water containing KBr/KBrO3 and hydroxylamine and reduction with SnCl2 in HCl using external calibration. The matrix interferences and the effect of nitrogen oxide vapors have been evaluated and the method validated using a certified reference material.
