Valorization of natural drug products : from extraction to encapsulation

2020

This thesis was carried out at the Laboratory of Pharmacognosy and the Laboratory of Pharmaceutical technology, at the UFR Sciences de Santé, circonscription Pharmacy, in the University of Burgundy Franche-Comté, to find new natural molecules to encapsulate. First of all, we focused on the natural saponins from plants of the Vietnam biodiversity and the french horticulture, belonging to the three genera Dracaena, Cordyline (Asparagaceae) and Weigela (Caprifoliaceae). The work led to the successful isolation and elucidation of 42 natural saponins using various chromatographic techniques. The structures were determined by ESI mass spectrometry and NMR spectroscopy. Among the 17 pure compounds…

Influence of solvent-free reticulation treatment on edible film properties based on chitosan and gelatin, containing or not natural antioxidants : ph…

2015

This thesis aims to develop an edible packaging made of a mixture of chitosan and gelatin (beef or fish), to better understand the influence of the electron beam irradiation and of the incorporation of natural antioxidants on the physico-chemical and functional properties of the films. A study of the effect of irradiation on antioxidants release kinetics in a simple liquid medium was studied for validation. A preliminary study first showed that the densification of the film-forming solution and the gel during drying does not depend on the thickness or concentration or time and suggests a Fickian diffusion of water in the matrix. The permeability to water vapor increases linearly with film t…

Selection of probiotic strains and improving the survival and functionality of an academic strain, Bifidobacterium bifidum, by changing the redox pot…

2012

The aim of this work was to select rationally a probiotic strain by setting up a screening method as well as study and understand the impact of the redox potential (Eh) and gas bubbling on the survival of Bifidobacterium bifidum in a fermented dairy product. In a first time, we have developed selection techniques of probiotic bacteria on the criteria of viability / vitality (flow cytometry analysis) and on the criteria of functionality (antioxidant). We were able to select strains of industrial interest as well as an academic strain, Bifidobacterium bifidum.Secondly, we have studied effect of modifying redox potential by gas bubbling on the survival of B. bifidum in a fermented dairy produc…

Alpha-lipoic acid: molecular mechanisms and therapeutic potential in diabetes.

2015

International audience; Diabetes is a chronic metabolic disease with a high prevalence worldwide. Diabetes and insulin resistance are associated with the development of cardiovascular and nervous diseases. The development of these disorders reflects complex pathological processes in which the oxidative stress caused by reactive oxygen species (ROS) and reactive nitrogen species (RNS) plays a pivotal role. It is widely accepted that diabetes impairs endothelial nitric oxide synthase (eNOS) activity and increases the production of ROS, thus resulting in diminished NO bioavailability and increased oxidative stress. Alpha-lipoic acid (LA) possesses beneficial effects both in the prevention and …
