Development of picoplankton during natural and enhanced mixing under late-winter ice

2014

We studied the development of autotrophic picophytoplankton and heterotrophic bacterioplankton during the tran- sition from winter ice cover to open water under natural and manipulated mixing conditions in eutrophic Lake Vesijarvi. During the melting of the snow and ice cover, a convection layer developed which eventually met the che- mocline at the interface between the oxic and anoxic water masses. However, in the years with mechanically enhanced mixing, the whole water column remained well oxygenated and the deepening of penetrative convection was facilitated. Stochastic variations in weather, primarily the thickness of the snow cover, likely determined the timing of picophytoplankton gr…

Seasonal shifts between autochthonous and allochthonous carbon contributions to zooplankton diets in a subarctic lake

2011

We addressed the extent to which autochthonous and allochthonous carbon sources contribute to zooplankton diet throughout the year in oligotrophic subarctic Lake Saanajarvi. Optical measures of dissolved organic matter (DOM) indicated high water discharge and associated terrestrial DOM during winter and low inputs in summer. Bulk particulate organic matter (POM) showed d13C values consistent with allochthonous inputs of DOM. The most positive POM d13C values (227%) occurred during winter, when heterotrophic bacteria and nanoflagellates peaked in abundance; the isotopically lighter autotrophic phytoplankton shifted the POM d13 Ct o229% in summer. The d13C values of zooplankton were up to 4% …

Reconstruction and analysis of genome-scale metabolic model of a photosynthetic bacterium

2010

Abstract Background Synechocystis sp. PCC6803 is a cyanobacterium considered as a candidate photo-biological production platform - an attractive cell factory capable of using CO2 and light as carbon and energy source, respectively. In order to enable efficient use of metabolic potential of Synechocystis sp. PCC6803, it is of importance to develop tools for uncovering stoichiometric and regulatory principles in the Synechocystis metabolic network. Results We report the most comprehensive metabolic model of Synechocystis sp. PCC6803 available, iSyn669, which includes 882 reactions, associated with 669 genes, and 790 metabolites. The model includes a detailed biomass equation which encompasses…

Epiphytic bacteria make an important contribution to heterotrophic bacterial production in a humic boreal lake

2017

Bacterial production (BP) in lakes has generally only been measured in the pelagic zone without accounting for littoral BP, and studies of BP at the whole-lake scale are very scarce. In the dystrophic humic lakes, which are common throughout the boreal region, low light penetration through water has been assumed to seriously limit available habitats for littoral organisms. However, many highly humic boreal lakes have extensive partly submerged vegetation around the lake perimeter which can provide well-lit substrata for highly productive epiphyton. We measured epiphytic BP on the littoral vegetation and pelagic BP in a small highly humic boreal lake in Finland during an open-water season an…

Novel Autotrophic Organisms Contribute Significantly to the Internal Carbon Cycling Potential of a Boreal Lake

2018

ABSTRACT Oxygen-stratified lakes are typical for the boreal zone and also a major source of greenhouse gas emissions in the region. Due to shallow light penetration, restricting the growth of phototrophic organisms, and large allochthonous organic carbon inputs from the catchment area, the lake metabolism is expected to be dominated by heterotrophic organisms. In this study, we test this assumption and show that the potential for autotrophic carbon fixation and internal carbon cycling is high throughout the water column. Further, we show that during the summer stratification carbon fixation can exceed respiration in a boreal lake even below the euphotic zone. Metagenome-assembled genomes an…

Comparison of biocides, allelopathic substances and UV-C as treatments for biofilm proliferation on heritage monuments

2018

Abstract UV-C and biocide treatments are frequently used to treat historical monuments contaminated by autotrophic biofilms. In this study, the authors compare for the first time the efficacy of these treatments against microorganisms such as cyanobacteria and algae proliferating in the Vicherey church (Vicherey, Vosges Department, France). To identify the most environmentally friendly and also efficient method, an allelopathic treatment was also tested. Colorimetric and physiological measurements of treated biofilms were thus monitored for 6 months. Fungi growing on necrotic matter from treated biofilms were sampled and sequenced. With biocides, results showed incomplete eradication of bio…

Molecular cause and functional impact of altered synaptic lipid signaling due to a prg‐1 gene SNP

2015

Loss of plasticity-related gene 1 (PRG-1), which regulates synaptic phospholipid signaling, leads to hyperexcitability via increased glutamate release altering excitation/inhibition (E/I) balance in cortical networks. A recently reported SNP in prg-1 (R345T/ mutPRG-1) affects ~5 million European and US citizens in a monoallelic variant. Our studies show that this mutation leads to a loss-of-PRG-1 function at the synapse due to its inability to control lysophosphatidic acid (LPA) levels via a cellular uptake mechanism which appears to depend on proper glycosylation altered by this SNP. PRG-1 +/ mice, which are animal correlates of human PRG-1 +/mut carriers, showed an altered cortical networ…

Nitrogen Loss from Pristine Carbonate-Rock Aquifers of the Hainich Critical Zone Exploratory (Germany) Is Primarily Driven by Chemolithoautotrophic A…

2017

Despite the high relevance of anaerobic ammonium oxidation (anammox) for nitrogen loss from marine systems, its relative importance compared to denitrification has less been studied in freshwater ecosystems, and our knowledge is especially scarce for groundwater. Surprisingly, phospholipid fatty acids (PLFA)-based studies identified zones with potentially active anammox bacteria within two superimposed pristine limestone aquifer assemblages of the Hainich Critical Zone Exploratory (CZE; Germany). We found anammox to contribute an estimated 83% to total nitrogen loss in suboxic groundwaters of these aquifer assemblages at rates of 3.5-4.7 nmol L -1 d -1, presumably favored over denitrificati…

Biological and chemical characterization of new isolated halophilic microorganisms from saltern ponds of Trapani, Sicily

2021

Abstract Halophilic microorganisms inhabiting hypersaline environments such as salt lakes, Dead Sea, or salt evaporation ponds, have acquired specific cell adaptation to grow within stressful conditions. In this study, we isolated heterotrophic and autotrophic microorganisms from several saltern ponds located at the Natural Reserve “Saline di Trapani e Paceco”, Sicily, Italy. The aim of the study was to investigate the biotechnological potential of new microbial strains from saltern ponds, by capturing their biological and chemical diversity. After the isolation and identification of the sampled strains, their growth capacity was determined under low and high salinity conditions. The metabo…
