Selection of Membrane RNA Aptamers to Amyloid Beta Peptide: Implications for Exosome-Based Antioxidant Strategies
2019
The distribution of amyloid beta peptide 42 (Aβ42) between model exosomal membranes and a buffer solution was measured. The model membranes contained liquid-ordered regions or phosphatidylserine. Results demonstrated that up to ca. 20% of amyloid peptide, generated in the plasma (or intracellular) membrane as a result of proteolytic cleavage of amyloid precursor proteins by β- and γ-secretases, can stay within the membrane milieu. The selection of RNA aptamers that bind to Aβ42 incorporated into phosphatidylserine-containing liposomal membranes was performed using the selection-amplification (SELEX) method. After eight selection cycles, the pool of RNA aptamers was isol…
Reproducibility of evoked and induced MEG responses to proprioceptive stimulation of the ankle joint
2022
Cortical processing of proprioceptive afference can be investigated by examining phase locked evoked and induced responses in cortical signals to passive movement stimuli. Reproducibility of evoked and induced responses has been studied using electroencephalography (EEG), but proprioceptive domain has received little attention. It is unclear whether evoked and induced responses to proprioceptive stimulation arising from the lower limbs are reproducible using magnetoencephalography (MEG). Nineteen healthy volunteers (18 right-foot dominant, 36.1 ± 6.6 yr, 7 females) were measured in two MEG sessions separated by 9 ± 5 days in which their right ankle was rotated intermittently using a pneumat…
Phase-space factors and half-life predictions for Majoron-emitting β−β− decay
2015
A complete calculation of phase space factors (PSFs) for Majoron-emitting 0νβ − β − decay modes is presented. The calculation makes use of exact Dirac wave functions with finite nuclear size and electron screening and includes lifetimes, single-electron spectra, summed electron spectra, and angular electron correlations. Combining these results with recent microscopic interacting boson model nuclear matrix elements (NMEs) we make half-life predictions for the ordinary Majoron decay (spectral index n=1 ). Furthermore, comparing theoretical predictions with the obtained experimental lower bounds for this decay mode we are able to set limits on the effective Majoron-neutrino coupling constant …
Alterations of panoramic radiomorphometric indices in children and adolescents with beta-thalassemia major: A fractal analysis study
2021
Background Beta-thalassemia major is an inherited disorder that can cause bone deformity and loss of bone mineral density. The objective of this study is to evaluate the cortical and trabecular mandibular bone morphology of children and adolescents who have beta-thalassemia major (ß-TM) using a fractal dimension (FD) analysis and different panoramic radiomorphometric indices with digital panoramic radiographic images (DPRIs). Material and Methods The study included 80 patients (with 40 patients each of ß-TM and control). The mandibular cortical width (MCW), panoramic mandibular index (PMI), mandibular cortical index (MCI), and simple visual estimation (SVE) were evaluated, and an FD analysi…
High-precision mass measurements for the isobaric multiplet mass equation atA= 52
2017
Masses of $^{52}$Co, $^{52}$Co$^m$, $^{52}$Fe, $^{52}$Fe$^m$, and $^{52}$Mn have been measured with the JYFLTRAP double Penning trap mass spectrometer. Of these, $^{52}$Co and $^{52}$Co$^m$ have been experimentally determined for the first time and found to be more bound than predicted by extrapolations. The isobaric multiplet mass equation for the $T=2$ quintet at $A=52$ has been studied employing the new mass values. No significant breakdown (beyond the $3\sigma$ level) of the quadratic form of the IMME was observed ($\chi^2/n=2.4$). The cubic coefficient was 6.0(32) keV ($\chi^2/n=1.1$). The excitation energies for the isomer and the $T=2$ isobaric analogue state in $^{52}$Co have been d…
Relative expression of cholesterol transport-related proteins and inflammation markers through the induction of 7-ketosterol-mediated stress in Caco-…
2013
Human diets contain sterol oxidation products that can induce cytotoxic effects, mainly caused by cholesterol oxides. However, phytosterol oxides effects have been less extensively investigated. This study evaluates the production of inflammatory biomarkers (IL-1β, IL-8, IL-10, TNFα) and the influence of gene expression transporters and enzymes related to cholesterol absorption and metabolism (NPC1L1, ABCG5/8, HMGCoA, ACAT) produced by 7-ketosterols (stigmasterol/cholesterol) in Caco-2 cells. These effects were linked to intracellular signaling pathways by using several inhibitors. Results showed 7-ketostigmasterol to have a greater proinflammatory potential than 7-ketocholesterol. In non-p…
Low cholesterol stimulates the nonamyloidogenic pathway by its effect on the α-secretase ADAM 10
2001
Biochemical, epidemiological, and genetic findings demonstrate a link between cholesterol levels, processing of the amyloid precursor protein (APP), and Alzheimer's disease. In the present report, we identify the α-secretase ADAM 10 ( a d isintegrin a nd m etalloprotease) as a major target of the cholesterol effects on APP metabolism. Treatment of various peripheral and neural cell lines with either the cholesterol-extracting agent methyl-β-cyclodextrin or the hydroxymethyl glutaryl-CoA reductase inhibitor lovastatin resulted in a drastic increase of secreted α-secretase cleaved soluble APP. This strong stimulatory effect was in the range obtained with phorbol esters and was further increa…

Multiple Resistance to Betalactam Antibiotics, Azithromycin or Moxifloxacin in Implant Associated Bacteria
2013
Background Antibiotics are more and more frequently prescribed in dentistry for prevention and treatment of oral diseases. Bacterial resistance to these agents is clearly increasing, including even previously susceptible micro-organisms and true pathogens. The aim of the present investigation was to examine resistant bacterial strains with respect to possible multiple antibiotic resistance. Methods In a previous investigation, implant-associated bacteria were tested first as mixed cultures and again as pure isolates (n = 138) for resistance to one of five antibiotics (ampicillin/AM, ampicillin + sulbactam/AB, azithromycin/AZ, penicillin/PG, moxifloxacin/MX) using the Etest. The resistance o…
