Methanogens play an important role in the global carbon cycle, because they are involved in the last step of anaerobic degradation of organic material to methane. Although the first report on methane emanation from aquatic muds was given by Alessandro Volta in the year 1776, the first methanogen was not obtained in pure culture before 1947. Special culture techniques had to be developed for growing the strict anaerobic methanogenic isolates. The methanogens were the first species of the archaeal domain (Archaea) detected. FT-IR spectroscopic investigation of bacterial cell envelopes from Zymomonas mobilis which have different surface hydrophobicities

Abstract Changes in the cell envelope composition of bacteria Zymomonas mobilis 113S which resulted in varied cell surface hydrophobicity (CSH) were examined using FT-IR-spectroscopy and conventional methods of biochemical analysis. Significant differences in all analyses were detected between hydrophilic and hydrophobic compounds in the envelopes of Z. mobilis . A significant positive linear relationship ( P Z. mobilis and the ratio of absorbance band intensities assigned to the CH 2 and CH 3 vibrations from lipids as well as the asymmetric and symmetric stretching vibrations of these methylene and methyl groups. SERS detection of cell surface and intracellular components of microorganisms using nano-aggregated Ag substrate

Abstract The intracellular and cell surface composition and structural features of gram-positive and gram-negative bacteria were identified using near-infrared surface-enhanced Raman scattering (SERS). The structural differences of components that reside in the cell envelope are manifested by their SERS spectra, e.g. gram-negative vs. gram-positive. Silver particles were used as a SERS substrate by exploiting the existence of strong local electromagnetic fields (hot spots) within nanoscale aggregates of the particles.
