In human osteosarcoma 3AB-OS cancer stem cells let-7d seems to have the dual function of oncogene/tumor-suppressor miRNA
2015
Introduction: Osteosarcoma (OS), an aggressive tumor a!ecting adolescents, shows therapy resistance and recurrence which can depend on cancer stem cells (CSCs), the source for tissue renewal and hold malignant potential; thus, OS treatment requires their eradication. Here, using 3AB-OS CSCs previously obtained from the OS-MG63 cells, we focused on the role of let-7d in managing stemness properties of 3AB-OS CSCs. Methods: 3AB-OS CSCs were stably transfected with pCDomH-plasmid, containing mir-let7-d, or empty vector as a control. Cell proliferation and viability were evaluated by EdU and Trypan blue assays, respectively. Sarcosphere and colony assays and wound healing and transwell invasion…
Rescue of ovarian function: stem cells and regenerative factors
2022
SICILIAN MANGO PEEL INDUCES CELLULAR STRESS ACCOMPANIED TO MITOCHONDRIAL DYSFUNCTION IN COLON CANCER CELLS
2021
Currently, cellular stresses as the oxidative, metabolic and genotoxic stress are considered the cause of many different human pathologies as neurodegenerative diseases (e.g.,Alzheimer’s, Parkinson’s, amyotrophic lateral sclerosis), alcoholic liver disease, chronic obstructive pulmonary disease, and also cancer. Although the role of cellular stress has been largely debated in cancer, nowadays some therapies aim to target the intracellular pro-oxidant/anti-oxidant balance triggering the tumor commitment to cell death. Therefore, it has become more necessary an improved understanding of cancer response to cellular stress that could be advantageous to develop cancer tailored therapies. In this…
Oxymetazoline modulates proinflammatory cytokines and the T‐cell stimulatory capacity of dendritic cells
2007
The nasal decongestant oxymetazoline (OMZ) is frequently used in the topical treatment of rhinitis/sinusitis. As proinflammatory cytokines play a critical role in the development and maintenance of local inflammation, the aim of this study was to investigate the influence of OMZ on immune cells in order to diminish the mucosal infiltration of the nose. Peripheral blood mononuclear cells (PBMC) from buffy coats of healthy volunteers were isolated and stimulated in the presence or absence of OMZ. In addition, monocyte-derived dendritic cells (DC) were generated and different concentrations of OMZ were added. DC phenotype and their T-cell stimulatory properties were analysed. The vasoactive su…
Pro-apoptotic activity of the phytochemical Indicaxanthin on colorectal carcinoma cells (Caco-2) and epigenetic CpG demethylation of the promoter and…
2012
Phytochemicals play prominent roles in human diet and nutrition as protective redo-active substances in prevention of several disorders and chronic diseases in humans. Today, their function as potent modulators of the mammalian epigenome-regulated gene expression is rapidly emerging. In the present study antiproliferative effects of Indicaxanthin (Ind) from the fruits of Opuntia ficus-indica (1), and potential influence on DNA methylation has been investigated on Caco-2 cells, a human cell line of colorectal carcinoma. Ind caused a clear dose- and time-dependent decrease of the cell proliferation (IC(50) 50 M) associated to apoptosis as demonstrated by phosphatidylserine externalization an…

Hepatocellular cancer cell lines, Hep-3B and Hep-G2 display the pleiotropic response to resveratrol and berberine
2022
Purpose Human carcinoma cells with different p53 status exposed to a combination of bioactive substances, resveratrol and berberine, revealed different responses in cell viability via p53-dependant apoptosis pathway activation. Materials and methods Using 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay, we investigated various and opposing effects in hepatocellular carcinoma cells, Hep-G2 and Hep-3B with different p53-status. Results Cells decreased in viability after treatment with dose-dependent concentrations of resveratrol and berberine. Hep-3B p53 mutants were more sensitive in comparison to the p53 wild type Hep-G2 cell line. A syne…
