A thoracic vertebral localization of a metastasized cutaneous Merkel cell carcinoma: Case report and review of literature

2017

Background: Merkel cell carcinoma (MCC) is a rare neuroendocrine skin tumor, which may be related to sun exposure. It can metastasize to lungs, liver and bone, leading to severe morbidity and mortality. Vertebral metastases from MCC are rare. The authors report the tenth case in the literature, a 59-year-old patient with MCC, which was primarily localized in the scalp, and later provoked distant metastasis to the thoracic spinal column. Case Description: A 59-year-old woman was admitted at our Unit of Neurosurgery with a 4-month history of progressive and severe dorsal back pain, without neurological signs. The patient had been surgically treated for a recidivated MCC in the occipital regio…

Higher resource level promotes virulence in an environmentally transmitted bacterial fish pathogen

2017

Diseases have become a primary constraint to sustainable aquaculture, but remarkably little attention has been paid to a broad class of pathogens: the opportunists. Opportunists often persist in the environment outside the host, and their pathogenic features are influenced by changes in the environment. To test how environmental nutrient levels influence virulence, we used strains of Flavobacterium columnare, an environmentally transmitted fish pathogen, to infect rainbow trout and zebra fish in two different nutrient concentrations. To separate the effects of dose and nutrients, we used three infective doses and studied the growth of bacteria in vitro. High nutrient concentration promoted …

Cooperation between CRISPR-Cas types enables adaptation in an RNA-targeting system

2020

AbstractCRISPR-Cas immune systems adapt to new threats by acquiring spacers from invading nucleic acids such as phage genomes. However, some CRISPR-Cas loci lack genes necessary for spacer acquisition, despite apparent variation in spacer content between strains. It has been suggested that such loci may use acquisition machinery from co-occurring CRISPR-Cas systems. Here, using a lytic dsDNA phage, we observe spacer acquisition in the native host Flavobacterium columnare that carries an acquisition-deficient subtype VI-B locus and a complete subtype II-C locus. We characterize acquisition events in both loci and show that the RNA-targeting VI-B locus acquires spacers in trans using acquisit…

Mucin induces CRISPR-Cas defence in an opportunistic pathogen

2021

AbstractParasitism by bacteriophages has led to the evolution of a variety of defense mechanisms in their host bacteria. However, it is unclear what factors lead to specific defenses being deployed upon phage infection. To explore this question, we exposed the bacterial fish pathogenFlavobacterium columnareto its virulent phage V156 in the presence of a eukaryotic host signal (mucin). All tested conditions led to some level of innate immunity, but the presence of mucin led to a dramatic increase in CRISPR spacer acquisition, especially in low nutrient conditions where over 60% of colonies had obtained at least one new spacer. Additionally, we show that the presence of a competitor bacterium…

Contributions to Branch-and-Price-and-Cut Algorithms for Routing Problems

2019

This article deals with new exact branch-and-price-and-cut algorithms for the solution of routing problems. Specialized methods for the pickup-and-delivery problem (PDP), the truck-and-trailer routing problem (TTRP), the periodic vehicle routing problem (PVRP) and a service network design and hub location problem (SNDHLP) are presented. We develop a new technique for the acceleration of bidirectional labeling algorithms by a dynamic choice of the merge point. Moreover, for variants of the PDP, the bidirectional labeling can be effectively applied for the first time. In the TTRP, we model the extension to a 2 days planning horizon and the consideration of a quantity-dependent transfer time. …

Branch-price-and-cut algorithms for the pickup and delivery problem with time windows and multiple stacks

2016

Abstract This paper proposes models and algorithms for the pickup and delivery vehicle routing problem with time windows and multiple stacks. Each stack is rear-loaded and is operated in a last-in-first-out (LIFO) fashion, meaning that when an item is picked up, it is positioned at the rear of a stack. An item can only be delivered if it is in that position. This problem arises in the transportation of heavy or dangerous material where unnecessary handling should be avoided, such as in the transportation of cars between car dealers and the transportation of livestock from farms to slaughterhouses. To solve this problem, we propose two different branch-price-and-cut algorithms. The first sol…

Stabilized branch-price-and-cut for the commodity-constrained split delivery vehicle routing problem

2019

Abstract In the commodity-constrained split delivery vehicle routing problem (C-SDVRP), customer demands are composed of sets of different commodities. The C-SDVRP asks for a minimum-distance set of routes such that all customer demands are met and vehicle capacities are respected. Moreover, whenever a commodity is delivered by a vehicle to a customer, the entire amount requested by this customer must be provided. Different commodities demanded by one customer, however, can be delivered by different vehicles. Thus, the C-SDVRP is a relaxation of the capacitated vehicle routing problem and a restriction of the split delivery vehicle routing problem. For its exact solution, we propose a branc…
