A Trans-amplifying RNA Vaccine Strategy for Induction of Potent Protective Immunity
2019
Here, we present a potent RNA vaccine approach based on a novel bipartite vector system using trans-amplifying RNA (taRNA). The vector cassette encoding the vaccine antigen originates from an alphaviral self-amplifying RNA (saRNA), from which the replicase was deleted to form a transreplicon. Replicase activity is provided in trans by a second molecule, either by a standard saRNA or an optimized non-replicating mRNA (nrRNA). The latter delivered 10- to 100-fold higher transreplicon expression than the former. Moreover, expression driven by the nrRNA-encoded replicase in the taRNA system was as efficient as in a conventional monopartite saRNA system. We show that the superiority of nrRNA- ov…
Towards efficient inductive synthesis: Rapid construction of local regularities
2006
Given several input/output examples of some function we can state the problem: what is the “simplest” function which complies with these examples. This problem is well studied and is known to be very hard in the general case. In this paper we address a special case of the problem, when the target function can be expressed as a simple composition of known functions. We propose a new inductive synthesis algorithm for this case and show that it is efficient enough to synthesize complex geometry formulas.
Generator-based PET radiopharmaceuticals for molecular imaging of tumours: on the way to THERANOSTICS.
2011
Generator-derived radionuclides for PET/CT imaging are promising for optimizing targeted radiotherapy by an individual patient-based approach, applying pre-therapeutic evaluation, as well as dosimetric calculations, and for measuring treatment response after radionuclide therapy.
Complex regional pain syndrome - phenotypic characteristics and potential biomarkers
2018
Complex regional pain syndrome (CRPS) is a pain condition that usually affects a single limb, often following an injury. The underlying pathophysiology seems to be complex and probably varies between patients. Clinical diagnosis is based on internationally agreed-upon criteria, which consider the reported symptoms, presence of signs and exclusion of alternative causes. Research into CRPS biomarkers to support patient stratification and improve diagnostic certainty is an important scientific focus, and recent progress in this area provides an opportunity for an up-to-date topical review of measurable disease-predictive, diagnostic and prognostic parameters. Clinical and biochemical attribute…
Optimal Tree Decompositions Revisited: A Simpler Linear-Time FPT Algorithm
2020
In 1996, Bodlaender showed the celebrated result that an optimal tree decomposition of a graph of bounded treewidth can be found in linear time. The algorithm is based on an algorithm of Bodlaender and Kloks that computes an optimal tree decomposition given a non-optimal tree decomposition of bounded width. Both algorithms, in particular the second, are hardly accessible. We present the second algorithm in a much simpler way in this paper and refer to an extended version for the first. In our description of the second algorithm, we start by explaining how all tree decompositions of subtrees defined by the nodes of the given tree decomposition can be enumerated. We group tree decompositions …
Visualization of Large Terrain Using Non-restricted Quadtree Triangulations
2004
This paper presents a set of new techniques oriented towards the real-time visualization of large terrains. These techniques are mainly focused on semi-regular triangulations of non-restricted quadtree terrain representations. Despite the fact that the paper shows that triangulations based on non-restricted quadtrees are as simple and efficient as those based on restricted quadtrees, the new triangulations avoid discontinuity problems among the boundaries of different patches without the need for tree balancing and extra triangles addition. Another important feature of the proposed triangulation is that it incorporates an efficient method for building triangle strips and triangle fans for t…
CF3: An Electron-Withdrawing Substituent for Aromatic Anion Acceptors? “Side-On” versus “On-Top” Binding of Halides
2016
The ability of multiple CF3 -substituted arenes to act as acceptors for anions is investigated. The results of quantum-chemical calculations show that a high degree of trifluoromethyl substitution at the aromatic ring results in a positive quadrupole moment. However, depending on the polarizability of the anion and on the substitution at the arene, three different modes of interaction, namely Meisenheimer complex, side-on hydrogen bonding, or anion-π interaction, can occur. Experimentally, the side-on as well as a η(2) -type π-complex are observed in the crystal, whereas in solution only side-on binding is found.
Synthesis and reactivity of new mono- and dinuclear niobium and tantalum imido complexes: X-ray crystal structure of [Ta(η5-C5H4SiMe3)Cl2{NC6Me4-4-(N…
2006
Abstract The reaction of [1,4-{SiMe3(H)N}2C6Me4] (1) with 2 equivalents of LiBun followed by the addition of SiMe3Cl gave the diamine compound [1,4-{(SiMe3)2N}2C6Me4] (2). [Ta(η5-C5H4SiMe3)Cl4] reacts with 2, in a 2:1 stoichiometric ratio, to initially yield a mixture of the dinuclear, [{Ta(η5-C5H4SiMe3)Cl2}2(μ-1,4-NC6Me4N)] (3), and mononuclear, [Ta(η5-C5H4SiMe3)Cl2{ NC6Me4-4-(N(SiMe3)2)}] (4), imido complexes. 3 can be obtained exclusively by submitting the reaction mixture to repeated cycles of evacuation, to remove volatiles, followed by addition of solvent and subsequent heating. The mononuclear imido complex 4 was isolated from the reaction of [Ta(η5-C5H4SiMe3)Cl4] with 2 in a 1:1 sto…
Hindering the formation of ferrocenes: mono(cyclopentadienyl)halo iron complexes [Fe(C5R5)X] containing a sterically bulky cyclopentadienyl ligand
2001
Abstract The reaction of lithium 1,2,4-tris(trimethylsilyl)cyclopentadienide Li(Si 3 Cp) with ferrous halide FeX 2 (X=Cl, Br, I) in THF at −95 °C gives a thermally extremely sensitive compound that can be regarded as a functional equivalent for the 14-electron fragment [Fe(Si 3 Cp)X]. Experiments to trap this species with TMEDA gives thermally sensitive, colorless, paramagnetic crystals of the composition [LiFe(Si 3 Cp)Cl 2 (TMEDA) 3 ]. Trimethylphosphite gives isolable 18-electron half-sandwich complexes [Fe(Si 3 Cp){P(OMe) 3 } 2 X]. [Fe(Si 3 Cp)X] is also formed by reducing the ferric derivative [Fe(Si 3 Cp)Cl 2 ] with zinc. Analogous reactivity towards ferrous halides is observed for the…
Prediction of lncRNA-Disease Associations from Tripartite Graphs
2021
The discovery of novel lncRNA-disease associations may provide valuable input to the understanding of disease mechanisms at lncRNA level, as well as to the detection of biomarkers for disease diagnosis, treatment, prognosis and prevention. Unfortunately, due to costs and time complexity, the number of possible disease-related lncRNAs verified by traditional biological experiments is very limited. Computational approaches for the prediction of potential disease-lncRNA associations can effectively decrease time and cost of biological experiments. We propose an approach for the prediction of lncRNA-disease associations based on neighborhood analysis performed on a tripartite graph, built upon …
