Development of a real-time PCR assay for detection and quantification of enterotoxigenic members of Bacillus cereus group in food samples

A highly sensitive real-time PCR (qPCR) procedure, targeting the phosphatidylcholine-specific phospholipase C gene (pc-plc), was developed for specific detection and quantification of strains belonging to Bacillus cereus group. The target region was selected based on the enterotoxigenic profiles of 75 Bacillus strains. The inclusivity and exclusivity of the RTi-PCR assay were assessed with 59 isolates of the B. cereus group, 16 other Bacillus spp., and 4 non-Bacillus strains. Humans may, therefore, be exposed to BPF, and an assessment of this risk is required. BPF has been shown to have genotoxic and endocrine-disruptor properties in a human hepatoma cell line (HepG2), which is a model system for studies of xenobiotic toxicity. Effect of ethanol on the ability of Oenococcus oeni to remove ochratoxin A in synthetic wine-like media

The study focused on the changes in bacterial population, malolactic fermentation and ochratoxin A (OTA) levels in cultures of Oenococcus oeni performed in synthetic medium supplemented with ethanol and OTA. Growth was better in cultures containing 5% ethanol and was not observed in cultures containing 15% ethanol. The OTA removal ability of O. oeni depended on ethanol and initial OTA levels. The highest toxin removal percentage was observed in cultures containing 5% ethanol and 2 μg OTA/l. We assessed the association between exposure to air pollution during pregnancy and anthropometric measures at birth in four areas within the Spanish Children’s Health and Environment (INMA) mother and child cohort study. Determination of total mercury in nuts at ultratrace level

Abstract Total mercury, at μg kg−1 level, was determined in different types of nuts (cashew nut, Brazil nuts, almond, pistachio, peanut, walnut) using a direct mercury analyser after previous sample defatting and by cold vapour atomic fluorescence spectrometry. There is not enough sensitivity in the second approach to determine Hg in previously digested samples due to the strong matrix effect. Mercury levels in 25 edible nut samples from Brazil and Spain were found in the range from 0.6 to 2.7 μg kg−1 by using the pyrolysis of sample after the extraction of the nut fat. Occurrence of Aflatoxins in Tigernuts and Their Beverages Commercialized in Spain

A method based on matrix solid phase dispersion extraction was applied to determine aflatoxins B(1), B(2), G(1), and G(2) from tigernuts and tigernut beverages. Recoveries of each aflatoxin from tigernut (spiked at 10 microg/kg level) and from tigernut beverages (spiked at 10 microg/L level) ranged from 72.3 to 82.1% and from 74.0 to 86.3%, respectively. The limits of quantification ranged from 0.21 to 1.49 microg/kg (for tigernuts) and from 0.13 to 0.57 microg/L (for tigernut beverages) studied using liquid chromatography with fluorescence detection.
