Identification of Cysteine Residues in Human Cationic Amino Acid Transporter hCAT-2A That Are Targets for Inhibition by N-Ethylmaleimide

In most cells, cationic amino acids such as l-arginine, l-lysine, and l-ornithine are transported by cationic (CAT) and y(+)L (y(+)LAT) amino acid transporters. In human erythrocytes, the cysteine-modifying agent N-ethylmaleimide (NEM) has been shown to inhibit system y(+) (most likely CAT-1), but not system y(+)L (Devés, R., Angelo, S., and Chávez, P. (1993) J. Physiol. 468, 753-766). We thus wondered if sensitivity to NEM distinguishes generally all CAT and y(+)LAT isoforms. Species-dependent Metabolism of Benzo[c]phenanthrene and Dibenzo[a, l]pyrene by Various CYP450 Isoforms

Abstract The metabolism of benzo[c]phenanthrene (B[c]Ph) and dibenzo[a, l]-pyrene (DB[a, l]P) with various CYP isoforms including rat 1A1, 1A2, 2B1, 2E1, human 1A1, 1A2, 1B1, 2A6, 3A4, 2E1 and fish 1A expressed in Chinese hamster V79 cells has been compared. Major differences in the catalytic activities and in the regioselectivity of the eleven CYP isoforms with B[c]Ph and DB[a, l]P as substrates have been observed. Role of cyclic nucleotide phosphodiesterase isoenzymes in contractile responses of denuded rat aorta related to various Ca 2+ sources

We have examined the cyclic nucleotide phosphodiesterase isoforms (PDE) involved in the contractile response of rat aorta to different agonists and different experimental procedures for use in functional studies. The inhibitory effect of AAL 05 on the different PDEs isolated from bovine aortic smooth muscle was examined. Compound AAL 05 appeared to be a selective PDE3 inhibitor. 3H-1,2-benzoxathiepine 2,2-dioxides: a new class of isoform-selective carbonic anhydrase inhibitors

Abstract A new chemotype with carbonic anhydrase (CA, EC 4.2.1.1) inhibitory action has been discovered, the homo-sulfocoumarins (3H-1,2-benzoxathiepine 2,2-dioxides) which have been designed considering the (sulfo)coumarins as lead molecules. An original synthetic strategy of a panel of such derivatives led to compounds with a unique inhibitory profile and very high selectivity for the inhibition of the tumour associated (CA IX/XII) over the cytosolic (CA I/II) isoforms. Electrochemistry of methylene blue at an alkanethiol modified electrode

International audience; Gold surfaces were derivatized with decanethiol. The electrochemistry of methylene blue at these modified electrodes was investigated in function of the gold cleaning process and compared with the results obtained at a bare gold electrode. Cyclic voltammetry at low methylene blue concentrations (c(MB) <= 16 mu M) yielded surface behavior data. The properties of the film vary with the electrode pretreatment. Without electrochemical cleaning step, the standard potential at a gold electrode modified with 1-decanethiol is nearly the same as on a bare gold electrode. Electrochemical response of ferrocenium filled calix[4]arene capsules in solution and immobilized on gold

Abstract Hydrogen-bonded dimers of tetra-urea calix[4]arenes are able to include ferrocenium ions as a guest. Heterodimeric capsules are exclusively formed in 1:1 mixtures of tetra-arylurea calix[4]arene 1 with tetra-tosylurea calix[4]arene 2. Sulfide groups attached via spacers on the urea rim of 1 allow their binding to metal surfaces rendering simultaneously their dissociation more difficult. Separation and identification of petroporphyrins extracted from the oil shales of Tarfaya: geochemical study

Abstract Vanadyl and nickel porphyrins were isolated from the oil shales of Tarfaya (Morocco) by extraction followed by column chromatography. The ratios and characteristics of these porphyrin complexes were essentially obtained on the basis of UV–visible and mass spectrometry data. Geochemical information could be drawn from these data. The nature and the contents of the metals coordinated and non-coordinated to porphyrin systems were also determined in this study.

Investigations on isomerization and rearrangement of polycyclic arenes under oxidative conditions – Anodic versus reagent-mediated reactions

Abstract Electro-organic conversions at an active molybdenum anode enable the formation of fused arenes. High chemoselectivity was achieved under anodic conditions, and a reagent-induced selectivity was observed by comparison with results of MoCl5-mediated reactions. Polycyclic arenes like phenanthrenes, triphenylenes, chrysenes, or helicenes were selectively obtained in yields up to 87% and in some cases unusual rearrangements were crucial for the product formation.
