Cyclam-strapped porphyrins and their iron(III)-copper(II) complexes as models for the resting state of cytochrome c oxidase
1999
International audience; The ESR study of two cyclam-strapped porphyrins in which, on one side, the cyclam is attached with a variable length linker to the porphyrin and, on the other side, a non-coordinating strap protects the iron from any intermolecular interaction, is reported. Variation of the linker length is made possible by the use of either a Michael reaction or a nucleophilic substitution, leading respectively to three or two carbon atom links. It is shown that in the case of the shortest link, the oxidized ironÈcopper complex exhibits a spin interaction. The distance between thetwo metal centers is evaluated to be around 4.5 Å, a value consistent with the one found in the natural …
Italian Peninsula preserves an evolutionary lineage of the fat dormouse Glis glis L. (Rodentia: Gliridae)
2010
The present study examines the population genetic structure of fifty-nine specimens of Glis glis (Linneaus, 1766) from thirteen localities in central Europe, sequencing a 400-bp segment of the mitochondrial cytochrome b (cyt b) gene and a 673-bp segment of the cytochrome c oxidase subunit I (COI) gene. The consensus tree obtained from Bayesian analysis revealed a robust dichotomy, showing two sister groups: one clade includes samples from a wide geographical area, extending from north-central Europe to northern Italy (major branch sensu Bilton), and the other comprises samples collected in central and southern Italy and in Sicily (Italian branch). According to the Tajima–Nei model, the two …
Estradiol deficiency and skeletal muscle apoptosis: Possible contribution of microRNAs.
2020
Background Menopause leads to estradiol (E2) deficiency that is associated with decreases in muscle mass and strength. Here we studied the effect of E2 deficiency on miR-signaling that targets apoptotic pathways. Methods C57BL6 mice were divided into control (normal estrous cycle, n = 8), OVX (E2 deficiency, n = 7) and OVX + E2 groups (E2-pellet, n = 4). Six weeks following the OVX surgery, mice were sacrificed and RNA isolated from gastrocnemius muscles. miR-profiles were studied with Next-generation sequencing (NGS) and candidate miRs verified using qPCR. The target proteins of the miRs were found using in silico analysis and measured at mRNA (qPCR) and protein levels (Western blot). Resu…
Identification of subspecies and parentage relationship by means of DNA fingerprinting in two exemplary of Pan troglodytes (Blumenbach, 1775) (Mammal…
2018
Four chimpanzee subspecies (Mammalia Hominidae) are commonly recognised: the Western Chimpanzee, P. troglodytes verus (Schwarz, 1934), the Nigeria-Cameroon Chimpanzee, P. troglodytes ellioti, the Central Chimpanzee, P. troglodytes troglodytes (Blumenbach, 1799), and the Eastern Chimpanzee, P. troglodytes schweinfurthii (Giglioli, 1872). Recent studies on mitochondrial DNA show the incorporation of P. troglodytes schweinfurthii in P. troglodytes troglodytes, suggesting the existence of only two subspecies: P. troglodytes troglodytes in Central and Eastern Africa and P. troglodytes verus-P. troglodytes ellioti in West Africa. The aim of the present study is twofold: first, to identify the cor…
The reduction of molecular oxygen by iron porphyrins
2002
Abstract Molecular assemblies have been synthesised to reproduce the structure of the cytochrome c oxidase (C c O) active site and to explore the roles played by its different features. It was discovered that a single iron porphyrin, adsorbed at the surface of a graphite electrode, is a selective catalyst for the four-electron reduction of dioxygen to water, at pH 7. Mössbauer spectroscopy on the reaction center of Rhodopseudomonas viridis
1992
Proteins called “reaction centers” (RC) can be isolated from many photosynthetic bacteria. They have one non-heme iron in a quinone acceptor region. The RC of Rhodopseudomonas viridis contains an additional tightly bound tetra-heme cytochrome c subunit. The electronic configuration of both cytochrome and the non-heme iron has been studied in the crystallized protein by Mossbauer spectroscopy at different redox potentials, pH-values, and with an addition of o-phenanthroline. At high potentials (Eh=+500mV) all heme irons are in the low spin Fe3+-state, and at low potential (Eh=−150mV) they are low spin Fe2+ with the same Mossbauer parameters for all hemes independent of pH. Redox titrations c…
A caspase-3-dependent pathway is predominantly activated by the excitotoxin pregnenolone sulfate and requires early and late cytochrome c release and…
2002
This study investigates the implication of mitochondria- and caspase-dependent pathways in the death of retinal neurones exposed to the neurosteroid pregnenolone sulfate (PS) shown to evoke apoptosis and contribute to amplification and propagation of excitotoxicity. After a brief PS challenge of intact retinas, caspase-3 and caspase-2 activation and cytochrome c release occur early and independent of changes in the oxidative state measured by superoxide dismutase activity. The temporal and spatial relationship of these events suggests that a caspase-3-dependent pathway is activated in response to cytochrome c release and requires caspase-2 activation and a late cytochrome c release in speci…
Histamine up-regulates phosphodiesterase 4 activity and reduces prostaglandin E2-inhibitory effects in human neutrophils.
2000
Objective: To investigate whether histamine produces up-regulation of phosphodiesterase (PDE) activity with functional consequences in human peripheral blood neutrophils.¶Methods: PDE activity was studied by a radioisotopic method following anion-exchange chromatography. Reverse transcriptase-polymerase chain reaction (RT-PCR) was used for detection of mRNA transcripts of PDE4 subtypes. Cyclic AMP (cAMP) levels were measured by enzyme-immunoassay, and superoxide generation by cytochrome c reduction.¶Treatment: Neutrophils were incubated for 4 h with histamine (1 μM).¶Results: PDE4 was the only isoenzyme activity increased in treated neutrophils. Kinetic analysis showed a ∼1.5-fold increase …
A new species of Rhopalosiphum (Hemiptera, Aphididae) on Chusquea tomentosa (Poaceae, Bambusoideae) from Costa Rica
2012
copyright 2012, Los autores y Zookeys. Datos incluidos por Lisela Moreira Carmona, responsable de depósitos del área de Patógenos y Plagas de Plantas (CIBCM-UCR). The new species Rhopalosiphum chusqueae Pérez Hidalgo & Villalobos Muller, is described from apterous viviparous females caught on Chusquea tomentosa in Cerro de la Muerte (Costa Rica). The identity of the species is supported both by the morphological features and by a molecular phylogenetic analysis based on a fragment of the mitochondrial DNA containing the 5’ region of the cytochrome c oxidase 1 (COI) and on the nuclear gene coding for the Elongation factor-1 alpha (EF1α). The taxonomic position of the new species is discussed…
