The in vivo use of optical imaging is still limited by the lack of near infrared emitting probes. This thesis work focuses on the optimization and valorization of a water-soluble fluorescent platform whose optical properties enable an in vivo use. Two distinct applications were investigated for this WazaBY (Water-soluble azaBODIPY) platform: use as a PET (or SPECT) / optical bimodal probe, and as a theranostic agent. Concerning the first project, we were able to develop a targetted SPECT/optical bimodal probe, which was radiometallated with indium 111. During the past 25 years the treatment of Wilms' tumors has been refined owing to a series of thoroughly performed clinical studies. The overall prognosis is currently 80% (long-term cures), but different therapeutic issues are still discussed controversially, especially the question of preoperative treatment. A number of different drugs have proved effective in the treatment of Wilms' tumors. It remains to be determined as to which drug combinations are the most effective for which histologic subtype and which stage of disease. A total of 62 patients at high risk for recurrence of superficial bladder cancer were selected for a study designed to compare the prophylactic efficacy of different doses and schedules of sequential intravesical instillations of epirubicin and interferon-alpha-2b and to evaluate which sequence could enhance the release of cytokines in the urine. Our investigations showed a significant increase in urinary concentrations of interleukins in patients who received the sequential intravesical administration of epirubicin and interferon-alpha-2b. International audience; Neurotensin receptor 1 (NTSR1) is overexpressed in most human pancreatic ductal adenocarcinomas. It makes it an attractive target for the development of pancreatic cancer imaging agents. In this study, we sought to develop a bimodal PET-fluorescent imaging agent capable of specifically targeting these receptors.
