Simple Summary Pancreatic cancer is a digestive tumor that is most difficult to treat and carries one of the worst prognoses. The anatomical location of the pancreas makes it very difficult to obtain enough tumor material to establish a molecular diagnosis, so knowing the biology of this tumor and implementing new targeted-therapies is still a pending issue. The use of liquid biopsy, a blood sample test to detect circulating-tumor DNA fragments (ctDNA), is key to overcoming this difficulty and improving the evolution of this tumor. DNA recovery from Droplet Digital™ PCR emulsions using liquid nitrogen

Droplet microfluidics is a technology that enables the production and manipulation of small volumes. In biosciences, the most popular application of this technology is Droplet Digital™ PCR (ddPCR™), where parallel nanoliter-scale PCR assays are used to provide a high sensitivity and specificity for DNA detection. However, the recovery of PCR products for downstream applications such as sequencing can be challenging due to the droplets' stability. Here we compared five methods for disrupting the droplets to recover DNA. Ekstracelulārās vezikulas (EVs) ir heterogēni, ar membrānu norobežoti pūslīši, kas satur dažādas makromolekulas, tai skaitā RNS. EVs izdala visu veidu organisma šūnas un tās atrodas visos cilvēka bioloģiskajos šķidrumos. Ir pierādīta to loma prostatas vēža (PV) attīstībā, progresijā un metastazēšanās procesos. Šajā darbā iekļauta astoņu potenciālo RNS biomarķieru analīze PV un labdabīgas prostatas hiperplāzijas (LPH) pacientu plazmas un urīna EVs, kā arī PV pacientiem pirms un pēc radikālas prostatektomijas, izmantojot pilienu digitālo PCR.
