Tooth size discrepancy in a Libyan population, a cross-sectional study in schoolchildren
2014
Objetives: The aim of this cross-sectional study was to investigate the tooth size discrepancy (TSD) in a group of Libyan schoolchildren, and to compare TSD between sexes. Material and Methods: The sample comprised 333 Libyan schoolchildren (162 males with a mean (SD) age of 14.4 (1.1) years, and 171 females with a mean age of 14.1 (1.1) years). Anterior and overall TSD ratios were computed using descriptive statistics. Sex differences were statistically assessed using an independent t-test ( P 0.05). The percentages of participants showing more than 2 SD variation for the anterior and overall ratios comprised 3% and 4.2% of the total sample, respectively. Conclusions: The anterior and over…
Thyroid Stimulating Hormone Receptor Antibodies in Thyroid Eye Disease-Methodology and Clinical Applications.
2018
Background Thyroid stimulating hormone receptor antibodies (TSHR-Ab) cause autoimmune hyperthyroidism and are prevalent in patients with related thyroid eye disease (TED). Purpose To provide a historical perspective on TSHR-Ab and to present evidence-based recommendations for clinical contemporary use. Methods The authors review the recent literature pertaining to TSHR-Ab in patients with TED and describe the various immunoassays currently used for detecting TSHR-Ab and their clinical applications. Results We provide a historical summary and description of the various methods used to detect TSHR-Ab, foremost, the functional TSHR-Ab. Increasing experimental and clinical data demonstrate the …
Magnetic separation of encapsulated islet cells labeled with superparamagnetic iron oxide nano particles.
2012
Islet cell transplantation is a promising option for the restoration of normal glucose homeostasis in patients with type 1 diabetes. Because graft volume is a crucial issue in islet transplantations for patients with diabetes, we evaluated a new method for increasing functional tissue yield in xenogeneic grafts of encapsulated islets. Islets were labeled with three different superparamagnetic iron oxide nano particles (SPIONs; dextran-coated SPION, siloxane-coated SPION, and heparin-coated SPION). Magnetic separation was performed to separate encapsulated islets from the empty capsules, and cell viability and function were tested. Islets labeled with 1000 μg Fe/ml dextran-coated SPIONs expe…

Bioassays for TSH Receptor Antibodies: Quo Vadis?
2015
Autoantibodies (Ab) to the TSH receptor (TSHR) are responsible for many of the clinical manifestations of Graves' disease (GD) and are specific biomarkers of this autoimmune thyroid disorder (AITD) [1,2,3]. These Ab can be measured either via competitive-binding immunoassays or with bioassays [4]. Antibody-binding assays only report the presence or absence of TSHR-Ab and their concentrations, but do not indicate their functional activity. Bioassays, in contrast, indicate whether TSHR-Ab have stimulatory or blocking activity [5]. Historically, bioassays for TSHR-Ab were research tools used to study the pathophysiology of GD. Recently, however, there are increasing data that demonstrate the c…

Binding Properties and Stability of the Ras-Association Domain of Rap1-GTP Interacting Adapter Molecule (RIAM)
2012
The Rap1-GTP interacting adapter protein (RIAM) is an important protein in Rap1-mediated integrin activation. By binding to both Rap1 GTPase and talin, RIAM recruits talin to the cell membrane, thus facilitating talin-dependent integrin activation. In this article, we studied the role of the RIAM Ras-association (RA) and pleckstrin-homology (PH) domains in the interaction with Rap1. We found that the RA domain was sufficient for GTP-dependent interaction with Rap1B, and the addition of the PH domain did not change the binding affinity. We also detected GTP-independent interaction of Rap1B with the N-terminus of RIAM. In addition, we found that the PH domain stabilized the RA domain both in …
Standardization of islet isolation outcome- A new automatic system to determine pancreatic islet viability
2011
Pancreatic islet transplantation is emerging as a therapeutic approach for patients affected by diabetes. This approach consists of a minimally invasive procedure replacing insulin-producing cells (pancreatic islets). The technique has been proven successful, but limitations have been identified. One of the major challenges of the procedure is the counting of the isolated pancreatic islets, which is currently jeopardized by subjectivity and inaccuracy. Determination of the accurate islet number is a crucial factor in determining the correlation between the isolation product and clinical outcome. In the proposed study, we have developed software capable of objectively evaluating islet number…
