ERYTHROCYTE CATALASE ACTIVITY IN EVER-SMOKERS AND NO-SMOKERS HEALTHY SUBJECTS: INFLUENCE OF FLAVONOIDS (QUERCETIN AND LUTEOLIN)

2013

ERYTHROCYTE CATALASE ACTIVITY IN EVER-SMOKERS AND NO-SMOKERS HEALTHY SUBJECTS: INFLUENCE OF FLAVONOIDS(QUERCETIN AND LUTEOLIN) Background. Oxygen free radicals are highly reactive species that can cause cell damage including lipid peroxidation, enzyme inactivation and DNA damage. Inactivation and removal of highly reactive species depend by the antioxidant defence systems. The catalase (CAT) belongs to the primary antioxidant defence system of the cell which catalyses the decomposition of hydrogen peroxide to water and molecular oxygen. Flavonoids are a group of polyphenolic compounds with different chemical structure and properties. They are widely distributed in fruits, vegetables, nuts, …

An affordable method to obtain cultured endothelial cells from peripheral blood

2013

The culture of endothelial progenitor cells (EPC) provides an excellent tool to research on EPC biology and vascular regeneration and vasculogenesis. The use of different protocols to obtain EPC cultures makes it difficult to obtain comparable results in different groups. This work offers a systematic comparison of the main variables of most commonly used protocols for EPC isolation, culture and functional evaluation. Peripheral blood samples from healthy individuals were recovered and mononuclear cells were cultured. Different recovery and culture conditions were tested: blood volume, blood anticoagulant, coating matrix and percentage of foetal bovine serum (FBS) in culture media. The succ…

Resveratrol reduces oxidative stress and cell death and increases mitochondrial antioxidants and XIAP in PC6.3-cells.

2010

Resveratrol, a polyphenol derived e.g. from red grapes, has been shown to mediate several positive biological actions such as protection of cells against oxidative stress. It can also influence cell signaling, but the mechanisms behind its antioxidant properties are largely unknown. Here we show that RSV reduces oxidative stress and enhances cell survival in PC6.3 cells depending on the concentration. In these cells, RSV increased the levels of antioxidants, SOD2 and TRX2, and of X chromosome-linked inhibitor of apoptosis protein. RSV also activated NFκB signaling as shown using luciferase reporter constructs. These findings show that RSV regulates oxidative stress and mitochondrial antioxi…

Physical, chemical and morphological changes of bread polyphenol extract on cellular cultures monolayer CaCo-2

2013

The effect on potential transepithelial electrics resistance (TEER) and the electronic scansion microscopy analysis (SEM) of CaCo-2 cells line monolayer could be one useful method to study the different antioxidant or pro-oxidant capacity values of different concentrations of the hydrolizated polyphenol extracts of bread made with wheat Sant’Agata cultivar (HBPE).The various concentrations of HBPE have been tested on culture in monolayer CaCo-2 cells seeded on polycarbonate filter, in the cell culture chamber (Ø 6.5 μm; area 0.33cm2; pore Ø 0.4 μm), at a density of 1.5x105 cells per filter and placed in a multiwells Falcon; the filter divided the chamber in apical and basal compartments tha…

An Investigation of the Seasonal Pattern of Mannitol Content in Deciduous and Evergreen Species of the Oleaceae Growing in Northern Sicily

2002

In several species of the Oleaceae, mannitol, already present at considerable levels, accumulates in response to stress. This family comprises both deciduous and evergreen species, and we investigated the role of mannitol in deciduous malacophyll and evergreen sclerophyll species growing under the same conditions in the field. The relationship between mannitol content and changes in rainfall or temperature was also studied. The mannitol content of leaves of Fraxinus ornus L., F. angustifolia Vahl., Olea europaea L. and Phillyrea media L. was determined by gas chromatography. Leaf samples were collected once a month for 1 year. In the two ash species, the seasonal pattern of mannitol content…
