Water vapor sensors based on the swelling of relief gelatin gratings

We report on a novel device to measure relative humidity. The sensor is based on surface diffraction gratings made of gelatin. This material swells and shrinks according to the content of water vapor in air. By sending a light beam to the grating, diffracted orders appear. Due to the gelatin swelling or shrinking, first order intensity changes according to the relative humidity. Calibration curves relating intensity versus relative humidity have been found. The fabrication process of diffraction gratings and the testing of the prototype sensing devices are described.

Mobilization of proline in the starchy endosperm of germinating barley grain.

In germinating grains of barley, Hordeum vulgare L. cv. Himalaya, free proline accumulated in the starchy endosperm during the period of rapid mobilization of reserve proteins. When starchy endosperms were separated from germinating grains and homogenized in a dilute buffer of pH 5 (the pH of the starchy endosperm), the liberation of proline continued in these suspensions. The process was completely inhibited by diisopropylfluorophosphate, indicating that it was totally dependent on serine carboxy-peptidases. A specific search for Vibrio vulnificus in natural marine samples from the Spanish Mediterranean Sea was carried out by nested PCR and cultural approaches using thiosulphate-citrate-bile salts-sucrose agar (TCBS) and cellobiose-polymixin B-colistin agar (CPC), incubated at 40 degrees C, as selective media. Presumptive colonies were identified by PCR using specific primers against 23S rRNA sequences.
