The cytoprotective protein MANF promotes neuronal survival independently from its role as a GRP78 cofactor
2021
Mesencephalic astrocyte-derived neurotrophic factor (MANF) is an endoplasmic reticulum (ER)-stress-regulated protein exhibiting cytoprotective properties through a poorly understood mechanism in various in vitro and in vivo models of neuronal and non-neuronal damage. Although initially characterized as a secreted neurotrophic factor for midbrain dopamine neurons, MANF has recently gained more interest for its intracellular role in regulating the ER homeostasis, including serving as a cofactor of the chaperone glucose-regulated protein 78 (GRP78). We aimed for a better understanding of the neuroprotective mechanisms of MANF. Here we show for the first time that MANF promotes the survival of …
Assembly of Spinach Chloroplast ATP Synthase Rotor Ring Protein-Lipid Complex
2019
Rotor ATPases are large multisubunit membrane protein complexes found in all kingdoms of life. The membrane parts of these ATPases include a ring-like assembly, so-called c-ring, consisting of several subunits c, plugged by a patch of phospholipids. In this report, we use a nature-inspired approach to model the assembly of the spinach (Spinacia oleracea) c14 ring protein-lipid complex, where partially assembled oligomers are pulled toward each other using a biasing potential. The resulting assemblies contain 23 to 26 encapsulated plug lipids, general position of which corresponds well to experimental maps. However, best fit to experimental data is achieved with 15 to 17 lipids inside the c-…
Molecular Mechanism of ATP Hydrolysis in an ABC Transporter
2018
Hydrolysis of nucleoside triphosphate (NTP) plays a key role for the function of many biomolecular systems. However, the chemistry of the catalytic reaction in terms of an atomic-level understanding of the structural, dynamic, and free energy changes associated with it often remains unknown. Here, we report the molecular mechanism of adenosine triphosphate (ATP) hydrolysis in the ATP-binding cassette (ABC) transporter BtuCD-F. Free energy profiles obtained from hybrid quantum mechanical/molecular mechanical (QM/MM) molecular dynamics (MD) simulations show that the hydrolysis reaction proceeds in a stepwise manner. First, nucleophilic attack of an activated lytic water molecule at the ATP γ-…
The Use of Ca- and Mg-Rich Fly Ash as a Chemical Precipitant in the Simultaneous Removal of Nitrogen and Phosphorus - Recycling and Reuse
2019
The European Union’s circular economy strategy aims to increase the recycling and re-use of products and waste materials. According to the strategy, the use of industry waste material should be more effective. A chemical precipitation method to simultaneously remove phosphorus and nitrogen from synthetic (NH4)2HPO4 solution and the liquid phase of anaerobic digestate using fly ash as a precipitant was tested. Fly ash is a waste material formed in the power plant process. It mainly contains calcium oxide (CaO) and magnesium oxide (MgO). Saturated precipitant solution was prepared from fly ash, which was added in small proportions to (NH4)2HPO4 solution during the experiment. Fly ash’s effect…

Measurement of pyrophosphate concentration in cells by simple terpyridine-Zn(II) probe
2015
Pyrophosphate (PPi) anion is an integral part of our physiology. It partakes in the crucial energy transactions in cells and proffers energy for the harmonious functioning of the cell machinery. Discrepancies in PPi concentrations have been associated with several pathological aberrations such as chronic pyrophosphate arthropathy, calcium pyrophosphate deposition disease, cancer etc. However, there still exists a strong demand for an efficient and specific cell based assay for PPi in order to tap its importance to full potential. Terpyridine-Zn2+ complex (ZnCl2L probe) binds with PPi in 1:3 stoichiometry. This association engenders an emission band (λem) at ~591 nm when excited at 440 nm wi…
