Dithiothreitol prevents age-associated decrease in oocyte/conceptus viability in vitro.

1998

The present study was designed to ascertain whether the negative effects on reproductive potential of post-ovulatory ageing in vitro of oocytes can be prevented by antioxidant therapy. Mouse metaphase II (MII) oocytes were aged in vitro for 12 h prior to insemination in the presence of varying concentrations of L-ascorbic acid, 6-methoxy-2,5,7,8-tetramethylchromane-2-carboxylic acid (Trolox), L-cystine dihydrochloride, ethylenediaminetetraacetic acid (EDTA), beta-mercaptoethanol and DL-dithiothreitol (DTT). In-vitro ageing of oocytes was associated with lower fertilization rate, higher proportion of concepti exhibiting cellular fragmentation at 24 h post-insemination and lower percentage of…

Antiapoptotic effect of calcitonin gene-related peptide on oxidative stress-induced injury in H9c2 cardiomyocytes via the RAMP1/CRLR complex.

2005

Calcitonin gene-related peptide (CGRP) plays an important role in the mediation of protective effects observed in situations such as ischemic preconditioning in rat hearts. In this study, we investigated in H9c2 rat cardiomyoblasts if the protective effect of CGRP could be linked to an inhibitory effect on the apoptotic pathway. We also determined the specificity of observed effects by treatment with adrenomedullin (ADM) in stress conditions generated by 100 microM hydrogen peroxide. Using MTT assays, we demonstrate that a pretreatment with CGRP decreases by half the loss of cell viability induced by H(2)O(2). CGRP inhibits phosphatidylserine externalization, caspase 3 activation and DNA fr…

Fragmentation of bile duct stones: a prospective systematic in vitro evaluation of argon plasma coagulation, cryotechnology, and water-jet technology

2009

Background and study aim Choledocholithiasis is a common disease in the West. Lithotripsy by mechanical methods using baskets and by laser or electrohydraulic methods varies in effectiveness. With argon plasma coagulation (APC), high temperatures are used for devitalization and fragmentation; cryogenic techniques use the selective controlled application of freeze-thaw cycles to devitalize pathological tissue; and the dissecting water jet exploits the high pressure action of a thin laminar jet. We aimed to assess the feasibility and effectiveness of APC, cryotechnology, and the dissecting water jet as options for the fragmentation of bile duct stones. Methods In an in vitro feasibility study…

DNA fragmentation index, pAKT and pERK1/2 in cumulus cells are related to oocyte competence in patients undergoing in vitro fertilization programme

2019

SummaryActivated pERK1/2 and pAKT are key players in supporting cell survival and proliferation pathways. Translocation of pERK1/2 into the nucleus, where it interacts with transcription factors and DNA itself, is instrumental in exerting an anti-apoptotic effect. In this study, pAKT levels, pERK1/2 nuclear localization and DNA fragmentation index (DFI) in cumulus cells of single cumulus–oocyte complexes of patients undergoing in vitro fertilization programmes were evaluated and correlated with the clinical outcome of the related embryos. For a positive clinical outcome of blastocyst development, pERK1/2 nuclear localization and DFI value had a significant inverse relationship, whereas the …

Hybrid nanocomposites with ultra-low filling content by nano-coating fragmentation

2021

In the current study, a new technique for the manufacturing of polymer nanocomposites (PNCs) with ultra-low filling content (<0.05 wt%) is presented. Specifically, this method has been validated through the production of polypropylene (PP) nanocomposites with Ag nanoparticles. The presented method first consisted in a metal nano-film deposition on PP pellet substrates by a physical vapor deposition (PVD) sputtering equipment. Subsequently, a percentage of the coated pellet was mixed to uncoated ones via an injection molding machine to produce the PNC in a single step. Different mixing percentages of coated PP pellet to the uncoated ones were studied. As a result, the fragmentation of the…

Mechanisms of cell death in canine parvovirus-infected cells provide intuitive insights to developing nanotools for medicine

2010

Jonna Nykky, Jenni E Tuusa, Sanna Kirjavainen, Matti Vuento, Leona GilbertNanoscience Center and Department of Biological and Environmental Science, University of Jyväskylä, FinlandAbstract: Viruses have great potential as nanotools in medicine for gene transfer, targeted gene delivery, and oncolytic cancer virotherapy. Here we have studied cell death mechanisms of canine parvovirus (CPV) to increase the knowledge on the CPV life cycle in order to facilitate the development of better parvovirus vectors. Morphological studies of CPV-infected Norden laboratory feline kidney (NLFK) cells and canine fibroma cells (A72) displayed characteristic apoptotic events. Apoptosis was f…

Mass measurements of As, Se, and Br nuclei, and their implication on the proton-neutron interaction strength toward the N=Z line

2021

Mass measurements of the $^{69}$As, $^{70,71}$Se and $^{71}$Br isotopes, produced via fragmentation of a $^{124}$Xe primary beam at the FRS at GSI, have been performed with the multiple-reflection time-of-flight mass spectrometer (MR-TOF-MS) of the FRS Ion Catcher with an unprecedented mass resolving power of almost 1,000,000. For the $^{69}$As isotope, this is the first direct mass measurement. A mass uncertainty of 22 keV was achieved with only 10 events. For the $^{70}$Se isotope, a mass uncertainty of 2.6 keV was obtained, corresponding to a relative accuracy of $\delta$m/m = 4.0$\times 10^{-8}$, with less than 500 events. The masses of the $^{71}$Se and $^{71}$Br isotopes were measured…

La fragmentation de l'orthographe française : le cas de Twitter

2018

The enormous development of new communication technologies, especially the Internet, causes an unprecedented reversal in our lives sińce the discovery of printer. Apart from their impact on interpersonal relationships, new communication technologies have also modified our writing practices. It is evident that the French language used in computer-mediated communication digresses more and more from the developed formal speeches, in particular by its “fragmented” orthographic form which approaches its o rai form. Although this “fragmented” writing is scorned by many educated people, these orthographic variants represent, according to us, the true state of the French language as used by native …
