Functional Implications of Multiple IM30 Oligomeric States
2019
The inner membrane-associated protein of 30 kDa (IM30), also known as the vesicle-inducing protein in plastids 1 (Vipp1), is essential for photo-autotrophic growth of cyanobacteria, algae and higher plants. While its exact function still remains largely elusive, it is commonly accepted that IM30 is crucially involved in thylakoid membrane biogenesis, stabilization and/or maintenance. A characteristic feature of IM30 is its intrinsic propensity to form large homo-oligomeric protein complexes. 15 years ago, it has been reported that these supercomplexes have a ring-shaped structure. However, the in vivo significance of these ring structures is not finally resolved yet and the formation of mor…
Citrus rootstock breeding: response of four allotetraploid somatic hybrids to Citrus tristeza virus induced infections
2018
Four allotetraploid somatic hybrids of citrus, with potential for rootstock improvement, have been evaluated for their response to Citrus tristeza virus (CTV) infection. CTV is the most important viral pathogen affecting citrus production worldwide. Somatic combinations of ‘Milam’ lemon (Citrus jambhiri Lush.) + Sour orange (C. aurantium L Osb.), Calamondin (C. madurensis Lour.) + ‘Keen’ sour orange (C. aurantium L.), Calamondin + ‘Femminello‘ lemon (C. limon L. Burm. F.) and Cleopatra mandarin (C. reshni Hort. ex Tan.) + ‘Femminello’ lemon, were studied. Plants were grafted with CTV-infected “Valencia” sweet orange budwood. Two different CTV strains collected in Sicily, considered as “mild…
Molecule structural factors influencing the loading of flavoring compounds in a natural-preformed capsule: Yeast cells
2016
International audience; Yeast cells are efficient microcapsules for the encapsulation of flavoring compounds. However, as they are preformed capsules, they have to be loaded with the active. Encapsulation efficiency is to a certain level correlated with LogP. In this study, the effect of structural factors on the encapsulation of amphiphilic flavors was investigated. Homological series of carboxylic acids, ethyl esters, lactones, alcohols and ketones were encapsulated into the yeast Yarrowia lipolytica. Although, in a single homological series, the length of the molecule and thus the LogP were correlated with encapsulation efficiency (EY%), big differences were observable between series. Fo…
The IM30/Vipp1 C-terminus associates with the lipid bilayer and modulates membrane fusion.
2017
IM30/Vipp1 proteins are crucial for thylakoid membrane biogenesis in chloroplasts and cyanobacteria. A characteristic C-terminal extension distinguishes these proteins from the homologous bacterial PspA proteins, and this extension has been discussed to be key for the IM30/Vipp1 activity. Here we report that the extension of the Synechocystis IM30 protein is indispensable, and argue that both, the N-terminal PspA-domain as well as the C-terminal extension are needed in order for the IM30 protein to conduct its in vivo function. In vitro, we show that the PspA-domain of IM30 is vital for stability/folding and oligomer formation of IM30 as well as for IM30-triggered membrane fusion. In contra…

Constitutive expression of clathrin hub hinders elicitor-induced clathrin-mediated endocytosis and defense gene expression in plant cells.
2012
International audience; Endocytosis has been recently implicated in the signaling network associated with the recognition of microbes by plants. In a previous study, we showed that the elicitor cryptogein was able to induce clathrin-mediated endocytosis (CME) in tobacco suspension cells. Herein, we investigate further the induced CME by means of a GFP-tagged clathrin light chain and a CME inhibitor, the hub domain of clathrin heavy chain. Hub constitutive expression does affect neither cell growth nor constitutive endocytosis but abolishes cryptogein-induced CME. Such an inhibition has no impact on early events in the cryptogein signaling pathway but reduces the expression of defense-associ…
Biotrickling filter modeling for styrene abatement. Part 1: Model development, calibration and validation on an industrial scale
2017
Abstract A three-phase dynamic mathematical model based on mass balances describing the main processes in biotrickling filtration: convection, mass transfer, diffusion, and biodegradation was calibrated and validated for the simulation of an industrial styrene-degrading biotrickling filter. The model considered the key features of the industrial operation of biotrickling filters: variable conditions of loading and intermittent irrigation. These features were included in the model switching from the mathematical description of periods with and without irrigation. Model equations were based on the mass balances describing the main processes in biotrickling filtration: convection, mass transfe…
Oxygen transmittance correction for solar-induced chlorophyll fluorescence measured on proximal sensing: Application to the NASA-GSFC fusion tower
2017
Since oxygen (O 2 ) absorption of light becomes more pronounced at higher pressure levels, even a few meters distance between the target and the sensor can strongly affect canopy-leaving Solar-Induced chlorophyll Fluorescence (SIF) retrievals. This study was conducted to quantify the consequent error propagation and the impact of ignoring oxygen absorption effects on proximal sensing SIF measurements based on the O 2 -A absorption band with field-acquired and simulated data. It was demonstrated that the uncorrected oxygen transmittance between target and sensor distance of 10 m can lead to SIF relative errors ranging from 66% to higher than 100% when using a Spectral Fitting (SF) technique …
Spatiotemporal patterns in methane flux and gas transfer velocity at low wind speeds: Implications for upscaling studies on small lakes
2016
Lakes contribute significantly to the global natural emissions of methane (CH4) and carbon dioxide. However, to accurately incorporate them into the continental carbon balance more detailed surveys of lacustrine greenhouse gas emissions are needed, especially in respect to spatiotemporal variability and to how this affects the upscaling of results. We investigated CH4 flux from a small, wind-shielded lake during 10 field trips over a 14 month period. We show that floating chambers may be used to calibrate the relationship between gas transfer velocity (k) and wind speed at 10 m height (U10) to the local system, in order to obtain more accurate estimates of diffusive CH4 flux than by applyin…
Lignin Inter-Diffusion Underlying Improved Mechanical Performance of Hot-Pressed Paper Webs
2021
Broader use of bio-based fibres in packaging becomes possible when the mechanical properties of fibre materials exceed those of conventional paperboard. Hot-pressing provides an efficient method to improve both the wet and dry strength of lignin-containing paper webs. Here we study varied pressing conditions for webs formed with thermomechanical pulp (TMP). The results are compared against similar data for a wide range of other fibre types. In addition to standard strength and structural measurements, we characterise the induced structural changes with X-ray microtomography and scanning electron microscopy. The wet strength generally increases monotonously up to a very high pressing tempera…
